Geomicrobiological approaches to immobilize radionuclides from the long term reservoirs of nuclear wastes has recently been attracted as a safe and sound nuclear waste management. Radionuclides are present as various oxidation states forming various compounds with naturally occurring minerals, organic and inorganic complexes. Based on recent reports related to safe disposal and sound management of radioactive waste, the interaction of microorganisms and radionuclides has been investigated through establishing KAERI underground research tunnel (KURT) in the Korea Atomic Energy Research Institute.As a pilot project, microbial reactions and capability of metal reduction using Fe(Ⅲ), Mn(Ⅲ/Ⅳ), Cr(Ⅵ), Se(Ⅵ) were examined under anaerobic condition.The purposes of this research were to investigate the diversity of microbes within KURT groundwater and to observe microbial metal reduction, and to analyze morphological structures and chemical composition of the microbes and the precipitates and its mineralogical composition. To cultivate metal reducing bacteria from groundwater samples of the KURT in S. Korea, different electron donors such as acetate, glucose, lactate, formate, pyruvate and Fe(Ⅲ)-citrate as an electron acceptor were added into growth media. The enriched cultures were identified by 16S rRNA gene sequence analysis for the diversity of microbial species.Using the microbes from enriched cultures, the effects of electron donors of acetate, glucose, lactate, formate, pyruvate and electron acceptors of akaganeite, manganese oxide, hausmannite, potassium dichromate, sodium selenate on microbial metal reduction were experimented respectively.SEM, TEM, EDX and XRD analyses were used to analyze morphological structure, chemical composition of microbes and crystallinity of the precipitates. Based on 16S rRNA gene analysis, 23 microbial species from the enriched cultures of KURT groundwater were identified. These microbial species are known to involve the degradation of organics, reduction and oxidation of metals including uranium and methane production. XRD and EDX analyses showed the bacteria reduced from ferric akagameite[FeOOH] to ferric and ferrous magnetite[Fe₃O₄], from tetravalent manganese oxide[MnO₂] to divalent rhodochrosite[MnCO₃], from trivalent hausmannite[Mn₃O₄] to divalent rhodochrosite[MnCO₃], from hexavalent potassium dichromate[K₂Cr₂O_(7)] to trivalent chromium and hexavalent sodium selenate[Na₂SeO₄] to selenium(0). The results showed that metabolic and respiratory activities of the microorganism under anaerobic condition caused biomineralization and immobilization of heavy metals. Therefore, the results of this study on microorganisms enriched from groundwater in KURT might be applicable to treat various heavy metal contaminants and even radionuclides from highly toxic, mobile to less toxic, immobile in subsurface environments. For safe disposal and sound management of radioactive waste, the ongoing investigation on long term behaviours and characteristics of radionuclides, microbial diversity and its interaction, and mechnisms of bioreaction of microgorganisms and radionuclides and other organic and inorganic matters should be understood in a whole system. The results of study could be useful as a pilot project.
최근에 발표된 주요한 연구결과들은 방사성폐기물처분장 내 방사성원소의 장기 거동에서 미생물의 역할에 대한 중요성을 강조하고 있다. 이에 본 연구의 목적은 고준위 폐기물 처리장 설립을 위한 기초연구로 지하처분연구시설(KURT; KEARI Underground Research Tunnel) 지하수 내에 존재하는 혐기성 미생물을 동정하고 이들 중 금속 환원 미생물을 농화 배양하여 미생물에 의한 Fe(Ⅲ), Mn(Ⅲ/Ⅳ), Cr(Ⅵ), Se(Ⅵ)의 지구미생물학적 환원에 대해 알아보는 것이다.미생물의 종 다양성을 알아보기 위해 16S rRNA 분석을 이용하여 KURT 지하수 내의 미생물과 농화배양 된 금속 환원 미생물을 동정하였다. 농화배양은 전자공여체로 acetate, glucose, lactate, formate, pyruvate와 전자수용체로 Fe(Ⅲ)-citrate를 주입한 혐기상태의 성장배지에 지하수를 주입하여 실시하였으며 배양된 금속 환원 미생물을 이용하여 akaganeite, manganese oxide, hausmannite, potassium dichromate, sodium selenate의 환원을 관찰하였다. 관찰은 SEM, TEM, EDX, XRD 분석을 실시하였다.16S rRNA 분석 결과 KURT 지하수 내에서는 총 23종의 미생물을 발견하였으며 이 미생물들은 유기물 분해 미생물, 금속 산화/환원 미생물, 메탄 생성 미생물, 우라늄 환원 미생물과 근연관계를 가지고 있음을 확인할 수 있었다. 또한 농화배양된 금속 환원 미생물은 Pseudomonas sp., Actinobacteria, Fusibacter paucivorans, Desulfuromonas michiganensis로 배양 조건에 따라 특정 미생물이 농화되었다. SEM, TEM, EDX, XRD 분석 결과 농화 배양된 금속 환원 미생물은 akaganeite(FeOOH)를 magnetite(Fe₃O₄)로, manganese oxide(MnO₂)와 hausmannite(Mn₂O₄)를 rhodochrosite (MnCO₃)로 환원 및 상전이를 일으켜 생광화작용을 발생시켰다. 또한 이온상태의 potassium dichromate(K₂Cr₂O_(7))를 Cr(III)으로, sodium selenate(NaSeO₄)를 Se(0)으로 환원 및 침전시켜 독성과 이동도가 낮은상태의 종으로 변화시켰다.본 연구결과로 볼 때 고준위 폐기물 지하처분연구시설인 KURT 지하수 내에는 다양한 종류의 금속 환원 미생물이 존재하고, 이들 금속 환원 미생물들은 Fe(Ⅲ), Mn(Ⅲ/IV), Cr(VI)와 Se(VI)을 환원 및 침전시킬 수 있으며, 이는 심부 지하의 혐기성 환경에서 핵물질 유출시 KURT 지하수 내의 금속 환원 미생물을 이용하여 방사능 원소 또한 직간접적으로 환원 및 침전시켜 핵물질의 확산을 방지시킬 수 있음을 시사한다.
