Neurological disorders such as Alzheimer's disease, Parkinson's disease, and stroke often result in a progressive decline in both memory and cognitive function. This neuropathological condition is caused by a neuroinflammatory response in the brain, and excessive expression of these neuroinflammatory cytokines causes neurotoxicity. As a result, non-steroidal anti-inflammatory drugs (NSAIDs) are being used as potential therapeutic agents in epidemiological and clinical studies. However, concerns about side effects coexist, and research on natural compounds with anti-neuroinflammatory properties is needed. In this study, I investigated the effect of an herbal mixture containing sage and St. John's Wort on cognitive and memory impairment. ‘Rosmarinic acid’ and ‘hyperoside’ were identified as index compounds that affect memory in Sage and St. John's Wort, respectively, using HPLC (high-performance liquid chromatography). In addition, the protective effect of the herbal mixture against oxidative stress induced by scopolamine and lipopolysaccharide (LPS) was confirmed using human neuroblastoma SH-SY5Y cells and murine microglial BV-2 cells. Cell viability was analyzed, nitric oxide (NO) levels were quantified, and inflammatory response cytokines; tumor necrosis factor-α (TNF-α), Interleukin 1β (IL-1β), and catalase production were quantified. As a result, a positive effect was confirmed for this parameter. Additionally, the herbal mixture was orally administered to mice and treated with scopolamine to induce an intentional inflammatory response and memory impairment. Cognitive and memory impairments were assessed using the Morris Water Maze and T-maze tests, and cytokines (TNF-α, IL-1β) and catalase in mouse serum were quantified. The group that received the herbal mixture at a concentration of 10mg/kg confirmed cognitive improvement effects in the Morris Water Maze and T-maze tests, and at the same time, the levels of TNF-α and IL-1β were significantly reduced and catalase expression was increased. These findings suggest that consumption of an herbal mixture containing sage and peppercorns may improve cognitive and memory disorders through anti-inflammatory and antioxidant mechanisms.
알츠하이머병, 파킨슨병, 뇌졸중과 같은 신경학적 장애는 종종 기억력과 인지 기능 모두 점진적 저하를 초래한다. 이러한 신경병리학적 상태는 뇌의 신경염증 반응으로 인해 발생하며, 이러한 신경염증성 사이토카인의 과도한 발현은 신경독성을 유발한다. 결과적으로, 비스테로이드성 항염증제 (NSAIDs)가 역학 및 임상 연구에서 잠재적인 치료제로 사용되고 있다. 하지만 부작용에 대한 우려가 공존하며, 항신경염증 특성을 지닌 천연 화합물의 연구가 필요하다. 본 연구에서는 세이지와 서양고추나물을 함유한 허브 혼합물이 인지 및 기억 장애에 미치는 영향을 연구했다. 세이지 (Sage)와 서양고추나물 (St. John's Wort)에서 각각 기억에 영향을 미치는 지표 성분으로 ‘rosmarinic acid’ 과 ‘hyperoside’를 HPLC (고성능 액체 크로마토그래피)를 사용하여 확인했다. 또한, 인간 신경모세포종 (SH-SY5Y)과 생쥐 소교세포 (BV-2)를 이용하여 각 스코폴라민과 LPS에 의해 유발된 산화 스트레스에 대한 한방 혼합물의 보호효과를 확인하였다. 세포 생존율 분석, 산화질소(NO) 수준 정량화, 염증반응 사이토카인 종양괴사인자 α (TNF-α)와 인터류킨 1β (IL-1β) 및 카탈라아제 생성량을 정량한 결과, 이러한 매개변수에 대한 허브 혼합물의 긍정적인 효과를 확인하였다. 또한, 허브 혼합물을 마우스에 경구 투여하고 스코폴라민을 처리하여 의도적인 염증 반응과 기억 장애를 유발했다. 인지 및 기억 장애는 Morris Water Maze 및 T-maze 테스트를 사용하여 평가되었으며, 마우스 혈청의 사이토카인 (TNF-α, IL-1β) 및 카탈라아제를 정량했다. 10 mg/kg 농도의 허브 혼합물을 투여 받은 그룹은 Morris Water Maze 및 T-maze 테스트에서 인지개선 효과를 확인하였고 동시에 TNF-α 및 IL-1β 수준이 크게 감소하였고 카탈라아제의 발현은 증가하였다. 이러한 발견은 세이지와 서양고추나물을 함유한 허브 혼합물의 섭취가 항염증 및 항산화 메커니즘을 통해 인지 및 기억 장애를 개선할 수 있음을 시사한다.
