It has been recently reported that 445-nm lasers have diverse effects on vascular contractility. Therefore, we decided to define the laser parameters for selective vasorelaxation and explore its potential mechanism. Using organ chamber arterial mechanics, nitric oxide (NO) assay, Matrigel assay, and microarray, we showed that a 200-Hz, 300-μs, 445-nm pulsed-laser (total energy of 600 mJ; spot size 4 mm) induced potent and selective vasorelaxation, without vasocontraction in rat mesentery artery. The laser stimulation increased NO production in the endothelial progenitor cells (CB-EPCs). The laser-induced vasorelaxation was inhibited by a non- selective NOS inhibitor, L-NAME. Microarray study in CB-EPCs suggested up-regulation of cryptochrome2 as well as nitric oxide synthase 1 and NOSTRIN (a NOS trafficker) by the laser. In conclusion, this study suggests that the 445-nm laser irradiation with specific parameters in frequency, and pulse duration (i.e., 200-Hz, and 300-µs, respectively) can be used for inducing regional vascular relaxation without vasocontraction possibly via the CRY photoreceptors and NOSs activation. This laser-induced control of regional blood vessel would be a potentially useful treatment for improving regional blood flow.
