Cohen syndrome (CS) is a rare autosomal recessive genetic disease with symptoms such as developmental delay, intellectual disability, microcephaly, hypotonia, abnormal facial features, high myopia and pulmonary retinal degeneration, and myopia. VPS13B gene mutation has been associated with CS. To understand the pathological mechanisms of CS, induced pluripotent stem cells were generated using fibroblasts of a patient with CS with novel mutations in the VPS13B gene. The cells were then differentiated into forebrain glutamatergic neurons and neurospheres. Transcriptome analysis of the iPSC-derived neurons that the synaptic-related functions were different from those of normal neurons and that many genes related to neurodevelopment were downregulated compared to people without CS. In addition, the activity of the autophagy pathway known to be important for neurodevelopment processes was impaired in the CS patient. Transcriptome analysis of the expression levels of genes related to autophagy revealed that the expression level of the ATG4C gene was significantly increased. The activity of autophagy was impaired in CS patient iPSC-derived glutamatergic neurons. To determine differences in the development process, neurospheres were generated. The proliferation of SOX2-positive cells was found to be reduced. Furthermore, the synaptic protein SV2B-positive small dots and neurite-like structures were significantly reduced in CS iPSC-derived glutamatergic neurons. Thus, synaptic-related functions, autophagy activity, and the proliferation of neural stem cells were impaired in CS-derived glutamatergic neurons and neurospheres. Accumulating studies have shown that dysfunctional autophagy is strongly associated with many neurodevelopmental disorders. To date, the overexpression of mammalian ATG8 has most commonly been used to monitor autophagy. However, due to artifacts in overexpression systems, there are some limitations. Therefore, new sensor was developed that could detect endogenous autophagosomes without overexpression or artifact problems. Using the LC3-interacting region (LIR) of proteins that interacts with mATG8s, and the hydrophobic domain (HyD) from Aplysia phosphodiesterase 4 (ApPDE4) that facilitates targeting of the cell membrane, a sensor was developed that could detect endogenous mATG8-positive autophagosomes. A total of 34 LIR motifs have already been identified. Among them, the LIR motif from the TP53INP2 gene was chosen to generate a HyD-LIR(TP)-GFP sensor that could detect all mATG8-positive autophagosomes. Specifically, it was found that the HyD-LIR (TP)-GFP sensor could detect more GABARAP and GABARAPL1-positive autophagosomes than the other sensors. HyD-LIR(TP)-GFP was also effectively located in endogenous mATG8-positive autophagosomes without affecting the basic autophagy activity. Moreover, it was confirmed that HyD-LIR(TP)-GFP could be used for live-imaging for the detection of dynamic autophagosomes. Although many LIR-containing proteins that could mediate the function of mATG8 have been identified, little is known about their physiological relevance to mouse behavior in a cell type-specific manner. To monitor and understand LIR-mediated mATG8 function in mice in vivo, knock-in mice expressing the HyD-LIR(TP)-Venus construct using the CaMKII-Cre inducible system was generated and validated as a mouse model for autophagy monitoring and studies. The expression of the HyD-LIR(TP)-Venus sensor was confirmed based on signals in the cerebral cortex and hippocampus, but not in the cerebellum. It was found that all mATG8-positive autophagosomes were consistent with the above data. In addition, it was confirmed that mATG8-positive autophagosomes accumulated in knock-in mice expressing the HyD-LIR (TP)-Venus sensor, thereby impairing autophagy activity. CLEM analysis revealed that late autophagosomes accumulated in the HyD-LIR(TP)-Venus expressing cells of knock-in mice. Later, through several experiments using mice with impaired autophagy activity in cortical and hippocampal neurons known to play a specific role in learning and memory, studies on how sensor-expressing mice can affect behavior related to autophagy are planned. Taken together, the CS iPSC model and the newly developed autophagy sensor will be very useful tools for future research on autophagy associated with many neurological.
코헨 신드롬은 드문 상염색체 열성 유전병으로 발달지연, 지적장애, 소두증, 근육긴장저하, 이상한 얼굴모습, 고도근시 및 맥랙망막퇴행위축, 그리고 과립 백혈구 감소증과 같은 증상이 나타나는 질병으로, 현재까지 VPS13B 유전자의 돌연변이가 일어나면 코헨신드롬이 발병한다고 보고되어 왔다. 이 VPS13B 유전자에 대해서는 아직 알려진 것이 많이 없지만, 소낭의 연관되는 단백질들을 분리하거나 운송하는데 중요한 역할을 한다고 알려져 있다. 하지만 코헨 신드롬 환자의 신경세포 내에서의 병리학적인 메커니즘은 알려진 바가 거의 없다. 따라서 이 학위논문에서는 코헨신드롬 환자의 세포 내의 병리학적인 메커니즘을 이해하기 위해 VPS13B의 새로운 돌연변이를 갖는 코헨신드롬 환자의 피부세포를 이용하여 유도만능줄기세포를 제작하였고, 이를 전뇌 흥분성 신경세포와 신경구로 분화를 시켜 실험을 진행하였다. 특히, 분화한 신경세포로부터 전사체 분석 방법을 이용하여 시냅스 연관 기능이 정상에 비해 달라져 있는 것을 확인하였고 특히나 신경발달에 관련된 많은 유전자들이 정상에 비해 낮게 조절되는 것을 보았다. 또한 신경발달과정에서 중요하게 작용하는 자가소화작용 경로 활성이 환자에게서 망가져 있는 것을 확인하였고, 전사체 분석에서 자가소화작용 관련 유전자들의 발현율을 관찰해 본 결과 ATG4C라는 유전자의 발현율이 매우 증가되어 있는 것을 발견하였고 이를 통해 자가소화작용의 활성도가 망가져 있다는 것을 발견하였다. 더욱이 SOX2-positive한 신경세포의 증식이 감소되어 있음을 보았고, 더욱이 시냅스 단백질인 SV2B-positive한 작은 점들과 신경돌기 유사 구조들이 상당히 환자의 신경세포에서 감소되어 있음을 볼 수 있었다. 이로써 코헨신드롬 환자의 신경세포에서 시냅스 관련된 기능들과 자가소화작용 기능, 신경줄기세포의 증식들이 망가져 있음을 확인할 수 있었다. 자가소화작용의 기능에 문제가 생기게 되면 많은 신경발달질환들에 연관된다는 연구들이 많아지고 있다. 현재까지 자가소화작용을 보기위해, mammalian ATG8의 과발현이 가장 흔하게 사용되어지고 있다. 하지만 과발현 시스템에는 여러 인위적인 문제들이 있기 때문에 한계점들이 존재한다. 그러므로 인위적인 문제없이 세포 내에 있는 자가소화작용 그대로를 볼 수 있는 새로운 센서를 개발하였다. 자가소화작용 핵심 유전자인 mATG8들과 상호작용하는 단백질들이 갖는 도메인 LC3-interacting region(LIR)과 세포막으로 가도록 도와주는 aplysia phosphodiesterase 4(ApPDE4)에서 유래된 hydrophobic domain(HyD)를 이용하여 자가포식체 막에 존재하는 mATG8들을 감지할 수 있는 센서를 개발하였다. 현재까지 알려진 LIR motifs 34개를 동정하였고 그 중 TP53INP2 유전자로부터 나온 LIR motif를 사용한 HyD-LIR(TP)-GFP가 모든 mATG8들에 positive한 자가포식체를 감지를 할 수 있음을 보았다. 그 중에서도 HyD-LIR(TP)-GFP 센서는 GABARAP과 GABARAPL1-positive 자가포식체를 더 많이 감지하는 것을 알게 되었다. HyD-LIR(TP)-GFP는 효과적으로 자가포식체에 위치하는 것을 보았고 이는 내재적인 자가소화작용의 활성도에 영향을 미치지 않는 것을 확인하였다. 더욱이 HyD-LIR(TP)-GFP가 dynamic한 자가포식체의 live-imaging을 할 수 있음도 확인하였다. 하지만 지금까지 많은 LIR 함유 단백질이 확인되면서 mATG8의 LIR 매개 기능을 갖는 것으로 생각되지만, 신경세포 특이적 상황에서 마우스 행동에 대한 생리적 관련성은 거의 알려지지 않았다. LIR 매개 mATG8 function을 보고 이해하기 위해 CaMKII-Cre inducible system을 이용하여 HyD-LIR(TP)-Venus를 forebrain에서 발현하는 knock-in mouse model을 만들었다. 이는 대뇌피질과 해마에서 발현이 됨을 확인하였고 이전 연구와 동일하게 mATG8s 단백질 모두를 감지할 수 있음을 확인하였다. 또한 센서를 발현시킨 마우스에서 mATG8s-positive 자가소포체가 축적되어 있는 것을 확인하였으며 자가소화작용 활성이 망가져 있음을 볼 수 있었다. 특이적으로 센서가 발현된 마우스의 신경세포에서 어떤 단계의 자가소포체가 영향을 받았는지를 확인하기 위해 CLEM 분석을 진행하였고 이를 통해 후기 자가소포체가 센서가 발현된 세포에서 축적되어 있음을 볼 수 있었다. 이후 여러 실험들을 통해 센서가 발현되어 자가소화작용의 활성이 학습과 기억 등을 담당하고 있는 대뇌피질과 해마의 신경세포에서 특이적으로 망가져 있는 마우스의 경우에 여러 행동들에 관련하여 어떠한 영향을 미치는지 알아볼 예정이다. 위의 실험 결과들을 통하여 코헨신드롬 유도만능줄기세포 모델과 새로 개발한 자가소화작용 센서는 앞으로 많은 신경질환들과 연관된 자가소화작용 연구를 위한 매우 유용한 도구가 될 것이다.
