Organic peroxides and hydroperoxides are known skin tumor promoters. Free radicals derived from these compounds are presumed to be the prominent mediators of tumor promotion. However, the molecular targets and action mechanisms of these species in tumor promotion are unclear. Phosphatase and tensin homologs deleted on chromosome 10 (PTEN) are tumor suppressors that play crucial roles in cell growth, cell survival, and proliferation by negative regulation of phosphoinositol-3-kinase/protein kinase B signaling (PI3K/AKT) pathway. PTEN is reversibly oxidized in various cells by exogenous and endogenous hydrogen peroxide. Oxidized PTEN is reversed by cellular reducing agents, among which thioredoxin (Trx) system plays a major role. Here, the role of cumene hydroperoxide (CuHP) on the redox state of PTEN was analyzed by using cells and in vitro assays. Recombinant PTEN was oxidized by CuHP. In contrast to H2O2, CuHP-induced PTEN oxidation was irreversible in HeLa cells. However, CuHP-oxidized PTEN was reduced by exogenous Trx system, indicating that the cellular Trx redox system for PTEN is impaired by CuHP. Taken together, these results demonstrate that CuHP induces PTEN oxidation and inhibits Trx system, which results in irreversible PTEN oxidation in HeLa cells. In addition, ablation of peroxiredoxin (Prdx) enhanced CuHP-induced PTEN oxidation in cells. Collectively, these results suggest a novel mechanism of CuHP in the promotion of tumorigenesis.Alcohol metabolism increases the production of reactive oxygen species (ROS), a major factor in the initiation and progression of alcoholic liver disease (ALD), which is characterized by abnormal lipid accumulation and liver injury. PTEN is a tumor suppressor that susceptible for oxidation-mediated inactivation, plays an important role in lipid metabolism and tumorigenesis in the liver. However, the exact molecular and cellular underlying mechanisms of ALD have not been well investigated. This study now shows that alcohol-induced ROS resulted in PTEN oxidation via formation of a disulfide bond between Cys124 and Cys71. Mechanistically, mitochondrial-derived ROS during alcohol metabolism contributes to PTEN oxidation. Thus, our findings identify PTEN is a critical intracellular target of oxidation by alcohol metabolism and provides a promising target for therapeutic intervention for ALD.

PTEN은 외/내인성 과산화수소에 의해 다양한 세포에서 가역적으로 산화된다. 산화 된 PTEN은 세포내 환원시스템에 의해다시 환원될 수 있는데 그 중에서도Trx 시스템이 중요한 역할을 한다.본 연구에서는, 세포와 체외 분석을 사용 하여 PTEN의 산화 환원 상태에 대한 CuHP의 영향을 분석했다. 재조합 PTEN을 CuH로 산화시켰더니, H2O2와 대조적으로, HeLa 세포에서CuHP-으로 유도된 PTEN 산화는비가역적이었다. 그러나, CuHP-에 의해 산화된 PTEN은 외인성 Trx 시스템에 의해 감소되었으며, 이는 PTEN에 대한 세포성 Trx 산화 환원 시스템이 CuHP에 의해 손상되었을 가능성을 암시한다.종합하면, 이들 결과는 CuHP가 PTEN 산화와 더불어 Trx 시스템을 억제하여 HeLa 세포에서 돌이킬 수없는 PTEN 산화를 초래한다는 것을 나타낸다. 또한, 퍼옥시레독신 (Prdx)의 제거는 세포에서 CuHP-유도된 PTEN의 산화를 향상시켰다. 종합적으로, 이 결과는 종양 형성의 촉진에서 CuHP의 새로운 메커니즘을 시사한다. 알코올 소비는 지질 축적 및 간 손상을 특징으로 하는 알코올성 간질환 (ALD)의 개시 및 진행의 주요 인자인 활성산소(ROS)의 생성을 증가시킨다.
