Part Ⅰ Separation of Epicatechin-3-Gallate(ECG) from Green tea by Enzymatic HydrolysisEpicatechin-3-Gallate (ECG), a polyphenol from green tea, has an anti-oxidant effect with chemo-preventive and chemo-therapeutic action. The separated ECG product from industrially extracted green tea contains much of Epigallocatechin-3-Gallate (EGCG) and other catechins. The ECG and EGCG are the difficult to separate by physical method because they have 2 and 3 hydroxyl group on the B-ring. We attempted to hydrolyze the EGCG, specifically using the lipase. After using more than 30 kinks of lipases, 10 kinds of solvents, and 4 kinds of buffers, we found that lipase 23 in methanol and in buffer(PH=10) condition can produce the best result of ECG ratio. The relative quantity of EGCG and ECG component mixture was increased from 30% to 87% detected by HPLC after the reaction.Part Ⅱ Enzymatic Hydrolysis Method to Get Pure 4-Carbon-Unit3-(benzyloxymethyl)oxirane-2ylmethanol composed of fully functionalized four carbon unit, is a crucial building block of biological active natural product. Enantiomerically pure compound may be regarded as a versatile intermediate for total synthesis goal via region- and stereoselective ring opening. As hydrolysis of ester compounds, PPL-catalyzed enzymatic reaction of hydrophobic substrate needs water and organic solvent. We chose 12 kinds of organic solvent to determine the distinction of PPL transformation in former three kinds of media. When it has been detected by HPLC after 3 hours and 48 hours, we can get the opposite enantiomer.
