Studies on the interactions of amyloidogenic proteins with trace metals, such as copper, have indicated that the metal ions perform a critical function in the early oligomerization process. In this study, we investigate the effects of Cu(II) ions on the active sequence regions of amyloidogenic proteins using mass spectrometry (MS) and collision induced dissociation tandem MS (CID-MS/MS). We chose three active sequence peptides NNQQNY, LYQLEN, and VQIVYK from yeast prion like protein Sup35, insulin chain A, and tau protein, respectively. The mass spectra indicate that Cu(II) ions bind to the three active sequence peptides. The tandem mass spectra of [Cu-dimer] complexes indicate that in NNQQNY Cu(II) preferentially binds to the N-terminal, while no such preference was suggested for LYLQEN or VQIVYK. Molecular dynamics simulations indicate greater stability of the N-terminal binding location for Cu(II) than the C-terminal.
아밀로이드 형성 단백질과 구리와 같은 미량금속과의 상호작용에 관한 연구에서 올리고머 형성과정에서 금속이온이 중요한 역할을 한다고 알려져 있다. 본 연구에서는 구리이온이 아밀로이드 형성 단백질의 중요 active sequence 부분에서 미치는 역할을 보고자 mass spectrometry (MS) 실험 및 collision induced dissociation tandem MS (CID-MS/MS)을 이용하였다. 효모 프리온 단백질 Sup35, insulin chain A, 및 tau protein에서 아밀로이드를 형성하기로 알려져 있는 NNQQNY, LYQLEN, VQIVYK 세가지 active sequence단백질들을 선택 하였다. MS spectra에서는 구리와 세가지 active sequence과의 상호작용이 관측이 되었고 MS/MS spectra에서는 구리가 NNQQNY에서는 N-terminal과의 상호작용을 선호하지만 LYQLEN과 VQIVYK에서는 선호도가 없다는 것이 관측되었다. Molecular dynamics simulation에서는 NNQQNY 단백질에서 N-terminal지역과 구리의 상호작용의 안정성을 보여준다.
