본 연구는 다양한 해산물과 해조류에 포함되어 있는 비소 화학종들의 분리 및 정량 분석 방법을 개발하는 데 중점을 두었다. 이를 위해서 해산물과 해조류에 포함되어 있는 비소 화학종들을 추출하기 위한 다양한 방법들과 비소 화학종들을 분리하기 위한 변수들을 조사하였다. 비소 화학종들의 정량 분석 방법을 개발하기 위해서 추출 방법, 분리 방법, 검출 방법 및 이와 관련된 기기적 변수와 방법적 변수들을 최적화하였다. 총 비소는 질산을 사용하여 두 가지 방법(쉐이킹 방법, 가열하는 방법)으로 추출한 후 유도결합 플라즈마 원자방출분광기 (ICP-AES)로 검출하였다. 해산물과 해조류에 포함되어 있는 비소 화학종들을 추출할 때 물과 메탄올의 혼합용액(1:1, V/V)을 추출 용매로 사용하여 초음파 처리하는 것이 최적의 조건이었다. 각 비소 화학종들을 이온크로마토그래피(IC)로 분리한 후 ICP-AES로 검출하였다. 본 연구에서 개발한 분석방법으로 해산물과 해조류 시료에 들어 있는 비소 화학종들의 정량분석에 적용하였을 때 비소 화학종들의 손실과 산화상태 변화가 없음을 확인하였고, 비소 화학종의 재현성 및 회수율이 좋았다. 본 연구에서 개발한 비소의 분석 방법을 적용하여 해산물(대게, 홍게, 문어, 낙지 및 오징어)과 해조류(김, 파래, 다시마, 미역 및 톳)에 포함되어 있는 비소 화학종들을 조사하였다. 모든 시료는 건조시킨 후 미세 분말로 만들어 사용하였다. 본 연구에서 사용한 해산물에는 주로 독성이 없는 arsenobetain (AsB)이 검출되었고 홍게에서 (152 ± 4) mg kg-1으로 가장 많이 검출되었으며, As3+, As5+, monomethylarsonicacid (MMA) 및 dimethylarsinic acid (DMA)와 같은 비소 화학종은 검출되지 않았다. 톳에는 주로 As5+가 존재하고 있으며, 그 농도는 34.0 ± 0.4 mg kg-1이였다. 파래, 다시마, 김 및 미역에는 As3+, As5+, MMA, DMA 및 AsB와 같은 비소 화학종이 존재하지 않았다.
This study was focused on the development of an analytical method for the separation and quantitation of arsenic (As) species in various seafoods and seaweeds. Various extraction methods for As species in seafoods and seaweeds were studied, and the various parameters for the separation of As species were also investigated. The developed method for the quantitative determination of As species included the selective extraction method, the optimization of the separation and detection method, and all the important instrumental and methodological parameters. The total As in each sample was extracted by using shaking and heating method in nitric acid medium. The total amount of As was determined by employing inductively coupled plasma-atomic emission spectrometry (ICP-AES). The best extraction method of As species was chosen with sonication in which a water : methanol mixture (1:1, v/v) was used as the extraction solvent. The As in each sample was found to exist in different oxidation state and the various forms of As species were separated by ion chromatography (IC), then detected by ICP-AES. The prevention of loss and transformation of As species was confirmed with the developed extraction and analytical method. Good reproducibility and recovery efficiency of As species with the selected extraction method were also obtained. In this study, seafoods (snow crab, red snow crab, octopus, octopus minor and squid) and seaweeds (red laver, green laver, sea tangle, sea mustard and hizikia) were investigated with the analytical method. All the samples were dried and ground to a fine powder. The main As species in seafoods was the non-toxic arsenobetain (AsB), and the highest concentration was found in the red snow crab (152 ± 4 mg kg-1, dry weight). Other As species (As3+, As5+, monomethylarsonic acid (MMA) and dimethylarsinic acid (DMA)) were not detected in seafoods. The major As species in a hizikia was As5+, and its concentration was 34.0 ± 0.4 mg kg-1 (dry weight). The As species (As3+, As5+, MMA, DMA and AsB) were not found in a green laver, sea tangle, red laver and sea mustard.
