There is considerable interest in the isolation of more potent anti-glycation compounds to treat diseases involving formation of advanced glycation end products (AGEs). Nonenzymatic protein glycation by reducing sugar, such as glucose or ribose, Maillard reaction is a complicated cascade of condensation, rearrangements, fragmentation, and oxidative modifications that lead to products collectively called advanced glycation end products(AGEs). The aim of this study was to investigate the activity compound in green marine algae Capsosiphon fulvenscens that responsible for the glycation inhibitory activity.The green marine algae Capsosiphon fulvenscens (CF) is a food supplement widely distributed in Korea, Japan coasts, with unique flavor and soft texture. In this study, 70% ethyl alcohol extract (CFE) and fractionated it with n-hexane (CFE-H), chloroform (CFE-C), ethyl acetate (CFE-C), butanol (CFE-B) and distilled water (CFE-D) was used to investigate the inhibitory activity of glycation. The hexane fraction (CFE-H), which has high inhibitory effect of AGEs showed the IC50 value as 205.4 μg/ml, was subjected to open column chromatography on silica gel with isocratic gradienting elution, hexane: ethyl acetate (55:45, v/v) as the elution. The collected 150 fractions then developed into 20 fractions with thin layer chromatography. With the result of the thin layer chromatography, collected the fractions with same spots, called CFE-H-A, CFE-H-B, CFE-H-C. The CFE-H-B is the highest active fraction. In anti-glycation effect, CFE-H- B (100 μg/ml) inhibited formation of 23.34% AGEs. For further purification of CFE-H-B, used sephadex LH20 open column and eluted with 100% MeOH. As above, developed the thin layer chromatography of the fractions according to the peak absorbance of UV-spectrum and two spots were obtained. PLC was used to obtain the single spot of CFE-H-B-1 and CFE-H-B-2. Analyzed them with HPLC, and the CFE-H-B-1 have the same peak with fraction 60. PLC was used to obtain the single spot about CFE-H-B-1 and CFE-H-B-2.
