본 연구는 다당류 기질로부터 다양한 올리고머 생산 및 당 전환을 위해 당 가수 분해 효소인 β-glucosidase를 생산하는 미생물을 분리하여 β-glucosidase 유전자 탐색 및 유용효소의 특성을 파악하고자 수행되었다. 새우 껍질로부터 β-glucosidase를 생산하는 능력이 우수한 균주 DAU5를 분리하였다. 이 균주의 16S rDNA 유전자의 염기서열을 분석한 결과, Exiguobacterium sp.과 높은 상동성을 나타내었으며, 이 균주를 Exiguobacterium sp. DAU5로 명명하였다. DAU5로부터 β-glucosidase 유전자는 shotgon 방법을 이용하여 획득하였다. 이 유전자의 염기서열 분석결과, 1,350bp, 450개의 아미노산으로 구성되어 있으며, SDS-PAGE에서 52kDa임을 확인하었다. DAU5로부터의 β-glucosidase와 다른 박테리아 유래의 β-glucosidase에서 존재하는 보존된 영역을 비교한 결과, glycoside hydrolase family 1에 속함을 확인하였다. 유용효소인 β-glucosidase의 특성을 파악하고자 발현벡터 pET-32a(+)에 유전자를 클로닝하였으며, Escherichia coli BL21(DE3)을 이용하여 발현하였다. His-fusion 정제 시스템에 의해 정제된 β-glucosidase는 pH 7.0과 45℃에서 최적의 활성을 나타내었다. 금속이온 Li^(+), K^(+), Zn²^(+), Mg²^(+), Na^(+), Ni^(+) 및 EDTA는 효소의 활성을 저해하였으며, Ca²^(+)는 효소 활성을 강하게 저해하였다. Cu²^(+)와 Hg²^(+)는 효소 활성에 대해 잠재적이 저해제임을 확인하였다. bglA의 기질 특이성을 조사한 결과, p-NPGlu와 p-NPGal에서 가장 높은 활성을 나타내었다. DAU5 균주로부터 생산된 β-glucosidase가 올리고당 분해함으로써 셀룰로오스 분해를 위한 발효 및 락토오스 올리고머의 생산에 유용할 것으로 사료된다.
A gram-negative bacterium, designated strain DAU5, was isolated from shrimp shell samples because it demonstrated high β-glucosidase activity. Through 16S rDNA gene sequence analysis the strain was identified as belonging to the genus Exiguobacterium. The β-glucosidase gene of Exiguobacterium sp. DAU5 was successfully cloned by the shotgun method. Nucleotide sequence determination by sodium dodecyl sulfate–ployacrylamide gel electrophoresis indicated that the gene for the enzyme contained 1350 bp, was coded by 450 amino acids, and was 52 kDa. The polypeptide exhibits significant homology with other bacterial β-glucosidases and belongs to the Glycoside Hydrolase Family 1. The β-glucosidase was purified by a His–fusion purification system. The optimal pH and temperature of the enzyme were 7.0 and 45 ℃, respectively. The enzyme activity was strongly inhibited by Ca2+, and Li+, K+, Zn2+, Mg2+, Na2+, Ni2+, and EDTA partially inhibited the enzyme activity. The BglA showed the highest activity with p-NPG and MUG. However, strain DAU5 β-glucosidase, which is responsible for degradation of oligosaccharides, is expected to be useful for the fermentation of cellulose degradation and the transglycosylation of saccharides.
