본 연구의 목적은 단백질의약품의 제형(formulation)에 계면활성제(surfactant) 로서 polysorbate80(PS80)과 poloxamer188(P188)이 병용되었을 경우 두 가지 물질을 동시에 분리 정량할 수 있는 분석법을 개발 및 검증(validation) 하는데 있다. 계면활성제는 단백질의 응집을 방지하고 interfacial stress를 감소시켜 단백질의 안정화에 달성하게 하여 제형에 부형제로 첨가되며, PS80과 P188은 가장 일반적인 계면활성제로서 쓰이고 있다. PS80의 분해 이슈와 P188의 visible particle 이슈를 감안하여 새로운 부형제 및 대안제를 평가하는 노력 외에도 PS80과 P188 두 가지 계면활성제를 잘 확립된 조합으로 병행 사용함으로서 보다 안정적인 제형을 개발하고 부작용을 최소화할 수 있다. 규제적 측면에서 ICH, CTD, FDA 및 EMA 규정에 기반하여 국내외 의약품 허가를 위해서는 부형제의 정량 분석법을 국제 기준에 적합한 방법으로 개발 및 검증하고, 적절한 규격을 설정하여 관리하여야 한다. 또한 출하시험 및 안정성시험으로 품질 평가하도록 해야한다. 따라서, PS80과 P188를 병행 사용 하였을 때 구조와 성질이 유사한 P188과 PS80을 분리 정량 할 수 있는 분석법을 개발하고자 하였다. 본 분석법 개발에서는 선행연구를 참고하여 HPLC (High Performance Liquid Chromatography, 고성능액체크로마토그래피) 분석 장비에 anion exchange와 reverse phase의 두 가지 mode가 결합된 mixed mode column 을 사용하였고, ELSD (Evaporative Light Scattering Detector, 증기광산란검출기)를 이용하였다. Acetone 침전법으로 완제의약품 중 API protein을 제거하고, calibration curve를 통해 PS80 및 P188을 정량하는 방법으로 개발하였다. 우선 PS80과 P188의 peak가 분리되어 특이적으로 검출되며, 간섭 peak가 없는 지를 확인하였다. 확인된 peak가 농도에 의존적으로 직선적인 측정값을 도출할 수 있는지 확인하여 정량시험의 가능성을 확인하였다. 이를 바탕으로 feasibility를 확인한 후, 참값에 근사한 값을 산출할 수 있는 분석법임을 확인하기 위해 회수율로서 정확성(accuracy)을 검증하고, 반복 시험 측정값의 편차를 통해 본 분석법의 정밀성(precision)을 검증하였다. 본 분석법으로 신뢰도 있게 확인할 수 있는 PS80 과 P188의 정량 범위(range)와 정량 한계 (limit of quantitation) 확인하였으며, 마지막으로 완건성(robustness)을 확인함으로서 column lot과 이동상 농도에 영향이 없음을 검증하였다. 해당 parametet를 검증하는 방법은 ICH Q2(R1) guideline에 따라 수행하였고, 국제 수준으로 검증 (validation)된 PS80 및 P188 정량 분석법 개발을 완료하였다. 이렇게 개발된 본 분석법은 국제공통기술문서(CTD) 기준 및 시험 방법에 적용할 수 있고 GMP 출하시험 항목으로 활용할 수 있다. 아울러, 부형제는 QbD 측면 중요 CQA로서 경시 변화 분석 (안정성 시험)에 사용할 수 있다.
The purpose of this study is to develop and validate an analysis method that can separate and quantify two surfactants at the same time when polysorbate80 (PS80) and polyoxamer188 (P188) are combined as surfactants in the formulation of protein drug product. Surfactants are added as excipients to protein drug formulations by preventing protein aggregation and reducing interfacial stress to achieve protein stabilization, and PS80 and P188 are used as the most common surfactants. In addition to efforts to evaluate new excipients and alternatives in consideration of the decomposition issue of PS80 and the visible particle issue of P188, more stable formulations can be developed and side effects can be minimized by using both PS80 and P188 surfactants in a well-established combination. In terms of regulation, ICH guidelines, CTD guidelines, FDA, and EMA regulations require that the excipient quantitative analysis method be developed and verified in a way that meets international standards, and appropriate standards are established and managed. In addition, it should be set as shipment test and stability test items to be evaluated for quality. Therefore, an analysis method capable of separately quantifying P188 and PS80 with different structures and properties is needed. In the development of this analysis method, a mixed mode column that combines two modes of anion exchange and reverse phase was used, and an evaporative light scattering detector (ELSD) was used. It was developed by removing API protein from finished drugs using Acetone precipitation and quantifying PS80 and P188 through calibration curves. First, the peaks of PS80 and P188 were separated and detected specifically, and it was confirmed that there was no interference peak. The possibility of a quantitative test was confirmed by confirming whether the identified peak could derive a linear measurement value depending on the concentration. Based on this, the accuracy was verified as a recovery rate to confirm that it is an analysis method that can calculate a value close to the true value, and the precision of this analysis method was verified through deviation of repeated test measurements. The quantitative range and limit of quantification of PS80 and P188, which can be reliably confirmed by this analysis, were confirmed, and finally, the column lot and mobile phase concentration were not affected by confirming robustness. The parameter of this method validation was confirmed according to ICH Q2(R1) guideline, and the development of the PS80 and P188 quantitative analysis method validated at the international level was completed. This analysis method developed in this way can be applied to the CTD and can be used as a GMP release test item. In addition, excipients can be used for time-dependent change analysis (stability test) as an important CQA in terms of QbD.
