외상성 사건과 같은 강력한 스트레스는 해마 의존성 기억을 손상시킨다. 기억을 담당하는 신경구조인 해마는 스트레스에 대한 반응 시스템인 시상하부–뇌하수체-부신 축을 통하여 글루코코르티코이드 분비를 조절한다. 해마는 많은 글루코코르티코이드 수용체를 가지고 있다. 그러므로 해마는 스트레스에 의해 분비된 글루코코르티코이드에 영향을 받고, 과도하게 분비된 글루코코르티코이드는 해마에 악영향을 준다. FK506 결합 단백질(FKBP5)은 글루코코르티코이드 수용체의 활성을 조절하는 HSP90의 공동 샤페론이다. 따라서, 스트레스에 대한 글루코코르티코이드 수용체의 신호 전달 및 인지 기억에 Fkbp5의 결실이 미치는 영향을 조사하기 위한 실험을 수행하였다. Fkbp5 유전자 결손 생쥐 및 야생형 대조 생쥐를 대상으로 피할 수 없는 무작위적 발바닥 전기 충격 스트레스를 준 후 코르티코스테론 수치, 해마의 인산화 글루코코르티코이드 수용체 수치 및 재인 기억을 측정하였다. 스트레스를 받은 Fkbp5 유전자 결실 생쥐는 스트레스를 받은 야생형 생쥐보다 체중 및 혈장 코르티코스테론 수준이 낮은 것으로 나타났다. Fkbp5 유전자 결손 생쥐는 스트레스 조건의 유무에 따른 인산화 글루코코르티코이드 수용체 양의 차이가 야생형 생쥐에서보다 증가하였다. 그러나, 유전자 결손 생쥐와 야생형 생쥐 사이의 재인 기억의 차이는 없었다. 이러한 결과는 Fkbp5 유전자 결손 생쥐가 시상하부-뇌하수체-부신 축과 해마의 글루코코르티코이드 수용체의 신호 전달을 더 효과적으로 조절함으로써, Fkbp5 유전자가 급성 스트레스에 대한 반응으로 스트레스 저항성과 연관이 있음을 시사한다.
Intense stress such as a traumatic event impairs hippocampus-dependent memory. A response system to stress is hypothalamic–pituitary–adrenal (HPA) axis and dysregulation of hippocampal glucocorticoid receptor (GR) implicates stress-induced memory impairments. FK506 binding protein (FKBP5) is a co-chaperone of HSP90 which regulates GR activity. Decreased expression levels of Fkbp5 are observed in the patients with post-traumatic stress disorder (PTSD). Therefore, the present experiment was conducted to examine the effects of Fkbp5 deletion on GR signaling and recognition memory under stressed condition. Fkbp5 knock-out (KO) and wild-type (WT) mice were subjected to periods of an unescapable unpredictable foot-shocks and then their corticosterone levels, hippocampal phosphorylated GR levels, and recognition memory were measured. The stressed Fkbp5 KO mice showed lower body weights and plasma corticosterone levels than the stressed WT mice. The Fkbp5 KO showed a greater difference in the pGR levels between no stress condition and stress condition than the WT mice. However, no differences in recognition memory between Fkbp5 KO and WT mice were observed. These results demonstrated that the Fkbp5 KO mice showed efficient negative HPA axis and hippocampal GR signaling, suggesting that Fkbp5 is associated with stress resilience in response to acute stress.
