Estrogen receptor is activated by the hormone estrogen, and controls the cell growth by altering gene expression as a transcription factor. So far two estrogen receptors have been founded: ERα and ERβ. Estrogen receptor is also implicated in development and progression of breast cancer. We found that ERα localized on the spindle and centrosome during mitosis. Depletion of ERα induced mitotic arrest at prometaphase and generated unaligned chromosome in metaphase cells. Furthermore, the level of β-tubulin and γ-tubulin is reduced in ERα depleted cells. Consistent with this, polymerization of microtubule in ERα depleted cells was decreased than control cells. We suggest that ERα regulates chromosome alignment and spindle dynamics by stabilizing microtubule during mitosis.
Estrogen receptor(ER)는 호르몬 estrogen에 의하여 활성화되고 transcription factor로써 유전자의 expression을 변화시켜 cell growth를 조절한다. 현재까지 ERα와 ERβ 두 개의 ER들이 발견 되였다. ER은 유방암의 발생과 진행에 영향을 준다고 알려져 있다. 우리는 ERα가 유사분열단계에서 spindle 과 centrosome에 위치하고 있다는 것을 발견하였다. ERα의 감소는 prometaphase에 arrest를 시키고 metaphase cell에서 unaligned chromosomes을 증가시킨다. 더 나가서 β-tubulin 과 γ-tubulin의 level은 ERα가 감소될 때 감소된다. 이 결과와 일치하게 MT의 polymerization은 ERα가 감소된 cell에서 감소하는 양상을 보인다. 그래서 우리는 ERα가 spindle microtubule을 안정화시켜 spindle dynamics와 chromosome의 alingment을 조절한다는 결론을 내리게 되였다.
