The need for new alternative energy is emerging to address problems and limitations of fossil fuels. Biorefinery industry producing bioenergy from biomass is sustainable, economical, and eco-friendly. Biomass is plant or animal materials used for energy production, and lignocellulosic biomass derived from plant dry matters is the most famous and abundantly available raw materials. Plant cell wall is mainly composed of lignocellulosic materials including cellulose, hemicellulose and lignin. Effective degradation of recalcitrant plant cell wall is important in the biofuel production process. For decomposition, a harsh environment using acids or bases at high temperatures is required. Therefore, in order to save time and money, it is desirable to use thermophiles that can degrade biomass and survive even in extreme environments. Sulfolobus is a species that can survive at high temperature that produce biofuels from biomass. Caldicellulosiruptor bescii has six representative multimodular glycoside hydrolases with two catalytic domains separated by one or multiple carbohydrate binding modules (CBM) to degrade plant cell walls. Among these enzymes, CelA is the most prevalent and important cellulase consisting of an N-terminal GH9 endoglucanase, three consecutive family 3 CBM, and a C-terminal GH48 exo-β-1,4-glucanase domain. This combination of endoglucanase and exoglucanase activity creates a synergistic effect on cellulose degradation. CelA from C. bescii was truncated in various ways and combined with the sso1354 gene in S. acidocaldarius. Since the signal peptide and the transmembrane domain derived from S. solfataricus were inserted, the fusion proteins were expressed in the cytoplasmic membrane and showed high activity in the acidic pH range. The introduction of GH48 exoglucanase enhanced the hydrolysis of cellulosic and hemicellulosic substrates due to its synergistic activity with GH12 endoglucanase. At this time, it was found that a proper spatial distance between exoglucanase and endoglucanase is important to show optimal enzymatic activity. The addition of GH48 domain and CBMs also conferred benefits to utilize crystalline cellulosic substrate which was not previously available. Therefore, our results indicate that the proper arrangement of endoglucanase, exoglucanase and CBM domain would be critical to degrade the lignocellulosic biomass effectively.
최근 예로부터 사용되어 왔던 화석연료의 고갈과 환경오염 문제로 인해 새로운 대체 에너지를 개발하고자 하는 움직임이 전세계적으로 일어나고 있다. 바이오매스로부터 에너지를 생산하는 바이오리파이너리 산업은 그러한 연구의 한 일환으로 친환경적이며 경제적이다. 바이오매스는 식물이나 동물 또는 그 부산물, 생활이나 산업부산물 등을 원료로 한다. 그 중에서도 가장 많이 알려진 것이 식물 유래의 목질계 바이오매스이다. 미생물을 이용한 바이오리파이너리 산업에서 효율적으로 연료를 생산해내기 위해서는 이러한 복합다당체 형태의 바이오매스를 잘 분해하는 것이 중요하다. 현재 바이오매스에서 바이오연료를 생산하는 단계는 고온, 산성 환경의 전처리 과정이 필요한데, 이를 위해서 고온 환경에서도 생존하며 효과적으로 바이오매스를 분해할 수 있는 균주를 선택한다면 전처리 과정의 비용을 절감할 수 있다. 그러한 균주 중 하나인 Sulfolobus는 고온, 산성 환경에서 생존하는 고세균이다. 이전 연구 결과에서 S. solfataricus는 바이오매스 분해와 관련된 효소가 많이 존재하고, S. acidocaldarius는 유전적으로 안정하여 산업적으로 많이 응용될 수 있다는 장점이 있기 때문에 이 두 장점을 합친 균주를 개발하였다. 그러나 이렇게 개발된 균주는 결정형 셀룰로오스는 잘 분해하지 못한다는 단점을 가지고 있었다. 이를 극복하기 위해 Caldicellulosiruptor bescii 유래의 CelA가 가지는 특성을 참고하여 새롭게 개량된 단백질을 발현시키는 균주를 개발하고자 하였다. CelA 단백질은 아미노 말단에 GH9 endoglucanase를 가지고 있고, 결정형 셀룰로오스와 결합하는 3개의 CBM3가 연속되며, 카르복실 말단에 GH48 exoglucanase를 가지고 있는 융합단백질 형태의 구조이다. 이를 참고하여 기존의 균주가 가지는 GH12 endoglucanase의 활성을 유지한 채, CBM3와 GH48 exoglucanase를 도입함으로써 바이오매스 이용능을 높인 균주를 개량하고자 하였다. C. bescii 유래의 CBM과 GH48 exoglucanase를 기존의 GH12 endoglucanase와 연결한 plasmid를 구축하였고, S. acidocaldarius에서 발현하는데 성공하였다. 발현된 융합단백질들은 세포막에서 생성되며 산성 pH범위에서 높은 활성을 보였으며, endoglucanase와 exoglucanase의 시너지 효과로 인해 셀룰로오스와 헤미셀룰로오스 기질을 더 잘 분해하는 효과를 나타내었다. 효소의 활성에는 exoglucanase와 endoglucanase의 적정한 공간적 거리가 중요하다는 것을 알 수 있었다. 결정형 기질에 대한 분해능을 높일 수 있는 CBM을 도입하여 기존에 잘 이용할 수 없었던 결정형 셀룰로오스를 이용하도록 균주의 범위를 확장하는 결과를 얻을 수 있었다. 결론적으로 목질계 바이오매스를 효율적으로 분해하기 위해서는 endoglucanase, exoglucanase 그리고 CBM의 적절한 배열이 중요하다는 것을 말해준다.
