The Caliciviridae family of RNA viruses currently includes 11 genera. The enteric caliciviruses Norovirus and Sapovirus are the most well-known members of this group are associated with severe gastroenteritis in both humans and animals. Porcine sapovirus cell entry has been discovered to be a complicated multistep process in which Porcine sapovirus surface protein sites interact successively with various cell surface molecules that act as attachment and entry receptors. Several caliciviruses have been found to use sialic acids or histoblood group antigens as attachment factors, as well as cell surface protein as receptors, such as CD300lf, CD300ld, and tight junction junctional adhesion molecule 1 (TJs). Occludin, a recently described molecule, is used as a coreceptor and interacts with the PSaV particles and has been found to be which via Rab5- and Rab7-dependent trafficking, travel in a complex into late endosomes. Viruses are obligatory parasites, many viruses enter and infect host cells by trying to hijack and stimulate host signaling systems. Even so the precise host signaling mechanisms used during Porcine sapovirus entry, on the other hand, are unknown. Infection with porcine sapovirus can result in epithelial tight junction (TJ) disruption, permeability of TJ increased due to reorganization of TJ proteins claudin, ZO-1, JAM-A, and intracellular translocation of occludin, among which act as coreceptors. Moreover, the cell signaling cascades or molecules that enable such processes remain a mystery. When polarized LLC-PK cells are infected with Porcine sapovirus, the RhoA, ROCK, and MLC signaling pathways are activated and causes TJ protein rearrangement in the cytoplasm, and especially the intracellular translocation of occludin, a reversible decrease of transepithelial resistance, increased paracellular permeability. Furthermore, lateral diffusion of the lipid probe, Bodify FL-C12-sphingomyelin-BSA complex, administered apically, revealed changes in TJ fence function in response to PSaV infection. Inhibiting this signaling pathway nullifies the effect of early Porcine sapovirus infection on TJ integrity and protein alteration. The activation of the RhoA/ROCK/MLC signaling pathway changes the distribution of TJ proteins and interrupts TJ integrity by contracting the actomyosin ring, allowing virions access to coreceptors and entry into cells. Cell death is a frequent consequence of virus infection. Three forms of cell death occur as a result of a viral infection: apoptosis, necroptosis, and pyroptosis. Caliciviruses have been shown in vitro and in vivo to induce intrinsic apoptosis., however, calicivirus-induced necroptosis remains to be fully elucidated. Necroptosis is a type of programmed cell death that is induced by RIPK1/RIPK3/MLKL signaling pathway. Infection of LLC-PK cells with porcine sapovirus (PSaV) Cowden strain as a representative of caliciviruses induces RIPK1-dependent necroptosis and acts as proviral compared to the antiviral function of PSaV-induced apoptosis. Furthermore, in virus-infected cells, PSaV-induced RIPK1-dependent necroptosis and apoptosis, which have proviral and antiviral effects, counterbalanced each other. This study could aid researchers in better understanding the nature of PSaV-induced necroptosis and apoptosis, as well as in the development of effective treatments for PSaV and other RNA virus infections.
바이러스는 숙주를 떠나서는 살 수 없는 기생충(obligatory parasites)이며, 많은 바이러스는 숙주 세포를 진입하고 증식하기 위해 숙주 세포의 신호전달체계를 이용하거나 활성화시킨다. Caliciviridae는 RNA 바이러스로서 현재 11개의 속을 가지고 있다. 이들 중 가장 주목받는 바이러스는 사포바이러스(SaV)와 노로바이러스(NoV)가 속해 있는 enteric calicivirus이며, 이들은 인간과 동물에서 중증 위장관염을 비롯 전신질환의 발생을 일으킬 수 있다. 돼지사포바이러스(PSaV)의 경우 감염 세포 내 진입 과정은 이 바이러스 표면 단백질에 의해 바이러스의 부착과 숙주 세포의 수용체와의 결합을 촉진하는 일련의 복잡한 과정을 통해 이루어지는 것으로 보고 있다. 그러나 현재 PSaV가 숙주 세포에 진입하기 위한 정확한 분자학적 빌셍 기전은 알려져 있지 않다. 최근 알려진 단백질인 occludin은 PSoV와 상호작용하는 co-receptor로 이용되며, 이 결합을 통해 Rab5 및 Rab7 의존성 세포 내 이동(trafficking)을 통해 말기 엔도좀 (late endosome)으로 이동할 수 있다고 한다. 또한, PSaV은 치밀이음 (tight junction, TJ) 단백질을 분리시키고 claudin, ZO-1, JAM-A 단백질의 위치를 재조정하여 TJ 간 투과성이 증가하도록 유도시킨다는 지견도 있었으나 이에 관련한 분자학적 세포 신호전달 체계 및 관여 단백질의 역할에 대해 완전히 구명이 미흡한 상황이다. 이에 이 연구에서는 polarized LLC-PK 세포에 PSaV를 감염시켜 RhoA/ ROCK/ MLC 신호전달 체계를 활성화시키는 것을 확인하였다. PSaV는 TJ 연관 단백질의 위치를 세포 내로 재배치시키는데, 특히 occludin의 세포 내 이동은 상피통과저항(transepithelial resistance)을 감소시키고, TJ 간 투과성을 증가를 발견하었다. 또한, PSaV 감염에 따른 TJ 결합부의 변화는 감염 세포 표면에 적용시킨 lipid probe인 Bodify FL-C12-sphingomyelin–BSA complex가 세포 사이 결합을 통과 및 확산되어 세포 벽면에 염색됨을 확인함으로써 이 신호전달체계를 재확인하였다. PSaV 감염 초기에 위 신호전달체계를 억제하면, TJ의 분리와 TJ 관련 단백질의 분포 변화를 막을 수 있었다. 이러한 결과로써 RhoA/ ROCK/ MLC 신호전달체계의 활성화는 TJ 관련 단백질 조성을 변화시키고, 액토미오신고리(actomyosin ring)의 수축을 방해함으로써, TJ 분리시키는데 이 때 바이러스가 공동수용체를 사용하여 세포 내로 진입할 수 있는 것으로 해석되었다.전반적으로 바이러스 감염 결과에 따른 세포 사멸은 다음 세가지 형태, 즉 apoptosis, necroptosis, pyroptosis 등의 세포사가 발생할 수 있다. Calicivirus는 동물의 세포에서 apoptosis을 스스로 유도할 수 있는 것으로 알려져 있으나, necroptosis에 관해서는 밝혀져 있지 않았다. Necroptosis는 프로그램화 된 세포괴사(programmed necrosis)로써 RIP 1 (receptor interacting protein kinase 1), RIP3 ((receptor interacting protein kinase 3), MLKL (mixed lineage kinase domain-like protein) 등과 같은 일련의 신호전달 단백질에 의해 조절된다. PSaV (Cowden strain)에 감염된 LLC-PK 세포의 본 실험의 결과를 보면, RIPK 1의존성 necroptosis는 프로바이러스 (proviral) 요소인 반면 PSaV에 의한 apoptosis는 항바이러스 (antiviral) 요소로 판명되었다. 결국 PSaV는 프로바이러스 과정으로 RIPK 1 의존적인 necroptosis와 항바이러스 반응인 apoptosis의 균형을 조절함으로써 세포 감염을 유도하는 것으로 해석되었다. 따라서 PSaV에 의한 apoptosis와 necroptosis의 병리학적 특성을 구명함으로써 PSaV뿐만 아니라 다른 calicivirus 속 바이러스에 대한 효율적인 진단과 치료의 수단을 찾는데 의미있는 단서가 될 수 있을 것이다.결론적으로 이 연구는 PSaV 의 숙주 세포에 대한 분자학적 감염 기전을 밝혀 내었고 더불어 이 병원체에 의해 발생된 주요 병변인 세포사들의 특성을 병리학적 및 분자병리학적 관찰을 통해 necroptosis에 대한 새로운 지견을 발견해 냄으로써 향후 PSaV 의 전반적인 기병론을 확립하는데 유용한 결과로 활용할 수 있을 것으로 기대한다.
