Microplastics (MPs) are persistent pollutants measuring less than 5 mm. They are widely distributed in aquatic and soil environments through various pathways. Among the various types of MPs, film-shaped MPs originating from packaging containers or agricultural mulching films have garnered relatively less attention compared to the prevalence of film-shaped MPs in aquatic and soil environments. Therefore, it is critical to determine their recovery rate in these contexts. Meanwhile, concerns about climate change are growing globally, and the impact of climate change on the environment is difficult to be predicted. Furthermore, if climate change affects the ecosystem in combination with other pollutants, it may become more challenging to assess their impacts on ecosystems. In Chapter 2, we analyzed the number of MPs and the recovery rate of film- shaped MPs in natural waters and soil. First, we analyzed the number of MPs in natural water samples. The experiment was conducted in the order of oxidation, density separation, and filtration. There were quite lots of MPs in natural water. After that, we analyzed recovery rate of film-shaped MPs in natural water based on their type and size, and in natural soil based on three different methods of detecting MPs. Polypropylene (PP), low-density polyethylene (LDPE), and polystyrene (PS) films were manually-cut using medical scissors and sorted using 500 μm and 850 μm sieves. Subsequently, 30 pieces of MPs categorized by type and size were mixed in natural waters. The recovery rate was calculated by quantifying the number of MPs on the filter paper under a fluorescence microscope. We found a significant positive correlation between MP size and the recovery rate: 79%, 90%, and 97%. The lower the density and the higher the hydrophobicity of MPs, the greater the recovery rate. In natural soil, the recovery rate of MPs was low, and there were significant differences in three different methods of detecting MPs. Therefore, it is crucial to recognize that when quantifying MPs in aquatic environments, the actual quantity of MPs exceeds the amount detected, and the extent of this underestimation depends on the type and size of MPs. Also, methods of detecting MPs in soil should be developed. In Chapter 3, the effects of film-shaped microplastics and increase in temperature on microbial communities were analyzed. Natural water and soils were placed in chambers with different temperatures to simulate warming. During the incubation experiment, extracellular enzyme activities, nitrogen mineralization, microbial biomass, and reactive oxygen species generation were analyzed. In water, extracellular enzyme activities had very low values, and there was no significant difference in extracellular enzyme activity according to temperature. Only in the case of BG enzymes, extracellular enzyme activity tended to decrease under MP treatment. The nitrogen mineralization was affected by high temperature, and when the temperature was high, the concentration of NH4 + tended to increase according to MP treatment. Reactive oxygen species were generated under both MP and temperature treatments. In soils, microbial biomass increased according to the increase in temperature and the concentration of MP treatments. Extracellular enzyme activities tended to decrease in response to the increase in temperature, and the addition of MPs induced no significant effects. The nitrogen mineralization was affected by high temperature in the initial stages of experiments. It was also found that these microbial responses were affected by the combined effects of MPs and increase in temperature. These results show that the combined effects of MPs and warming may differ from their individual impacts on microbial communities and highlight the importance of considering the combined effects of environmental pollutants and warming to secure water and soil health.
미세플라스틱(MPs)은 5 mm 미만의 오염물질로, 다양한 경로를 통해 수계 및 토양에 유입되어 광범위하게 분포하고 있다. 여러 모양의 MPs 중에서도 포장용기나 농업용 멀칭 필름에서 유래한 필름 형태의 MPs은 수계 및 토양에서 많이 나타나지만, 이에 대한 관심은 상대적으로 적다. 한편, 기후 변화에 대한 우려가 전 세계적으로 증가하고 있는 가운데, 기후 변화가 환경에 미치는 영향은 예측하기 어렵다. 더욱이 기후 변화가 다른 오염물질과 함께 생태계에 영향을 미칠 경우, 이러한 영향을 평가하는 것은 더욱 어려워질 수 있다. 2 장에서는 자연수에서 MPs을 검출하고, 자연수와 토양에서 필름 형태의 MPs의 회수율을 분석했다. 먼저 자연수에서 MPs의 농도를 분석했는데 이 실험은 산화, 밀도 분리, 그리고 여과의 순서대로 진행되었다. 그 후, MPs의 종류와 크기를 기반으로 자연수에서의 필름 형태 MPs의 회수율을 분석하였다. 폴리프로필렌(PP), 저밀도 폴리에틸렌(LDPE), 폴리스티렌(PS) 필름을 의료용 가위로 자르고 500 μm 및 850 μm 체로 걸렀다. 이후 종류와 크기별로 분류된 30개의 MPs 조각을 자연수에 혼합하였다. 회수율은 형광 현미경 아래에서 필터 위의 MPs 수를 측정함으로써 계산되었다. MPs 크기와 회수율 간에 유의한 양의 상관관계가 있었고, 밀도가 낮을수록 그리고 소수성이 클수록 회수율이 증가하는 경향이 있었다. 토양에서의 회수율은 검출 방법에 따라서 달랐다. 따라서 수계에서 MPs를 검출할 때 검출량은 과소평가될 수 있기 때문에 실제 존재량이 더 클 수도 있고, 이 과소평가의 정도는 MPs의 종류 및 크기에 따라 다를 수 있다. 토양에서의 미세플라스틱 검출법은 추가 연구가 필요할 것으로 보인다. 3 장에서는 필름 형태의 MPs 및 온난화가 수계 및 토양 미생물에 미치는 영향을 분석하였다. 자연수와 토양 시료를 온난화 처리 챔버에 두고 다양한 농도의 MPs를 처리하고 배양했다. 배양 실험 기간 동안 효소 활성도, 질소 무기화, 미생물 생체량 및 산화 스트레스를 분석하였다. 수계 미생물은 효소 활성도가 매우 낮았고, 온도에 따라 효소 활성도에 유의한 차이가 없었다. BG 효소의 경우에만 MPs 처리가 수행될 때 효소 활성도가 감소하는 경향이 있었다. 질소 무기화는 MPs의 영향을 받지 않았으며, 온도가 높을 때 NH4+ 농도가 증가하는 경향이 있었다. 산화 스트레스는 MPs 와 온난화의 상호작용에 의한 영향을 받은 것으로 나타났다. 토양에서는 MPs와 온난화에 의해서 미생물 생체량이 증가하는 경향이 있었다. 또한 효소 활성도는 온난화에 의해서 감소하고, 질소 무기화는 실험 초기에만 MPs와 온난화의 영향을 받았다. 또한 이러한 미생물 반응은 MPs 와 온난화의 상호작용에 의한 영향을 받은 것으로 나타났다. 본 연구의 결과는 MPs 및 온난화의 복합작용이 수계 및 토양 미생물에게 미치는 영향이 MPs와 온난화가 독립적으로 미치는 영향과는 다를 수 있으며, 추후 연구에서도 환경 오염물질과 온난화의 복합 작용을 고려하는 것이 중요함을 시사한다.
