[Introduction]Previously, we have found that autophagy activated MI stage in porcine oocytes during in vitro maturation. In this study, we tried to investigate relationship between autophagy and oocyte maturation. [Materials and Methods]In order to confirm media effect for autophagy activation, we compared autophagic activation by LC3-II detection at 0 h, 14 h, 28 h, 42 h in each TCM199 and NCSU-23 media during in vitro porcine oocytes maturation. Next, we measured LC3-II by westernblot after inhibiting nuclear maturation of oocytes by cAMP treatment of full time in vitro culture while control group with cAMP treatment of 22 h in vitro culture, in order to clarify that nuclear maturation affect to autophagy. In other hands, we counted matured oocytes after inhibiting autophagy by treatment with wortmannin or mixture of E64d and PepstatinA during 42 h of in vitro culture. [Results] In result, both media showed same autophagy pattern which LC3-II amount represent maximum at 14 h then gradually reduced. About the experiment of cAMP treatment, both groups of cAMP which was applied for different treatment time showed same amount of LC3-II while their maturation rate was represented to approximately four times higher in half time of cAMP treatment group than full time of cAMP treatment group. In other hands, autophagy inhibition during in vitro maturation by wortmannin treatment decreased oocytes maturation rates approximately half although autophagy inhibition by E64d and PepstatinA mixture did not affect to oocytes maturation rate. Overally, we suggest that oocytes maturation do not exist up-stream of autophagy but autophagy have possibility to be existed at up-stream of oocyte maturation.
