目的:探讨脂质体和厚朴酚联合热疗对人腺样囊性癌肿瘤模型的增殖抑制作用。方法用 DOTAP:Chol 脂质体包裹和厚朴酚来进行抗肿瘤体内实验;构建小鼠人腺样囊性癌肿瘤模型,瘤内注射和厚朴酚并联合局部热疗,观察肿瘤大小及小鼠生存期及生长状态,对移植瘤进行 CD31免疫组化染色和 TUNEL 分析。结果脂质体和厚朴酚联合热疗的抗肿瘤效应更强,联合治疗组肿瘤的体积明显小于对照组、热疗组、和厚朴酚组(P <0.05),部分肿瘤完全消退。联合治疗组小鼠的生存期有明显获益(P <0.05),在残存肿瘤中,肿瘤组织内微血管密度低于其他组,凋亡细胞明显增多。结论脂质体和厚朴酚能明显抑制人腺样囊性癌增殖,与热疗联合能够增强诱导肿瘤细胞凋亡,产生更强肿瘤杀伤效应。
Objective To assess the inhibitory effects of liposomal honokiol on proliferation in human adenoid cystic carcinoma in vivo. Methods DOTAP-embedded honokiol was adopted and injected in animal tumor models of adenoid cystic carcinoma(ACC-2cells) cells.The mice were treated with 0.9% saline,local thermotherapy,lipo-honokiol,and lipo-honokiol plus thermotherapy respectively. Tumor size,suvival time,adverse events were observed.Histological analyses of tumors were taken.Results We found that lipo-honoki-ol plus thermotherapy increased the apoptotic tumor cells dramatically with smallest mean tumor volumes,microvessel density and surviv-al time.Conclusions This study suggests that lipo-honokiol may suppress the proliferation of adenoid cystic carcinoma cells.The com-bined treatment of lipo-honokiol plus thermotherapy may enhance the apoptosis process in adenoid cystic carcinoma(ACC-2cells)cells.
