Dr. Zümrüt Ögel Eylül 2002, 100 sayfa Bacillus subtilis'te kompetans gelişimi, lipopeptid antibiyotik sürfaktin biyosentezi ve sporulasyonun indüklenmesi `quorum-sensing` (hücre yoğunluğunun algılanması) global regülasyon mekanizması ile kontrol edilmektedir. B. subtilis'in belirli suşları tarafından sentezlenen ve bir dipeptid antibiyotik olan basilisin biyo sentezinin de yine aynı mekanizmanın kontrolü altında gerçekleştiği kısa süre önce laboratuvarımız tarafından rapor edilmişti B. subtilis'tG iki hücre dışı sinyal peptidi, ComX feromonu ve CSF (Kompetans ve Sporulasyon Faktörü) ComA transkripsiyon faktörünü kontrolederek kompetans gelişimi ve sürfaktin biyosentezini sağlamaktadır. Önceki çalışmamızda phrC, com A ve opp genlerinin bloke edildiği mutantlar oluşturulmuş ve dipeptid antibiyotik basilisin biyosentezinin ComA, PhrC ve SpoOK (Opp) kontrolü altında gerçekleştiği gösterilmişti. Şimdiki çalışma, bahsedilen regülasyon mekanizmasında var olan diğer genetik belirleyicilerin basilisin biyosenteziyle olan muhtemel ilişkilerini aydınlatmayı amaçlamıştır. Bu amaç çerçevesinde, öncelikli olarak srfA operonu ve comP, comQ, abrB, spoOA, spoOH, comS ve srfB genlerinin insersiyonel inaktivasyonları (kaset mutaj enezleri) yapılmış, bunun için basilisin üreticisi suş B. subtilis PY79 ilgili genlerin bloke mutantlarının kromozomal DNA'larıyla transforme edilmiştir. Daha sonra seçilen transformantlarda basilisin üretimindeki değişiklikler `kağıt diski-agar difüzyon` metodu ile incelenmiştir. ComQ-ComX ve ComP'nin bacilysin biyosentezi için gerekli olduğu bulunmuştur. srfA operonunun inaktivasyonu basilisin üretme kabiliyetinin yok olmasına neden olmuş, bu da sürfaktin operonunun basilisin biyosentezinde doğrudan fonksiyonu olduğunu kanıtlamıştır. abrB geninin spoOA mutantlarında bloke edilmesi spoOA mutasyonunun neden olduğu basilisin- negatif fenotipi suprese etmiş, aynı genin basilisin üreticisi suş PY79'da bloke edilmesi ise basilisin üretimini yaklaşık iki kat arttırmıştır. Prof. Dr. Zümrüt Ögel September, 2002, 100 pages In Bacillus subtilis, competence development, lipopeptide antibiotic surfactin biosynthesis and sporulation initiation are controlled by `quorum-sensing` (cell density signaling) global regulation mechanism. Biosynthesis of dipeptide antibiotic bacilysin produced by certain strains of B. subtilis was recently reported by our laboratory to be under the control of the same mechanism. In B. subtilis, two extracellular signaling peptides, ComX pheromone and CSF (Competence and Sporulation Factor) stimulate 111the development of genetic competence and surfactin biosynthesis in response to high cell density (quorum-sensing) by regulating the activity of transcription factor ComA In our previous study, we have shown that biosynthesis of dipeptide antibiotic bacilysin is linked to ComA and PhrC (CSF) in a SpoOK (Opp)-dependent manner by constructing phrC-, comA- and o/?p4-disrupted mutants of B. subtilis. The objective of the present research is to elucidate the possible relationships between the other genetic determinants involved in this control system and bacilysin biosynthesis. In view of this objective, insertional inactivations via cassette mutagenesis of certain genetic components of quorum-sensing global regulation, namely srfA operon, comP, comQ, abrB, spoOA, spoOH, comS and srfB genes were made by transforming bacilysin producer strain B. subtilis PY79 with chromosomal DNAs from the blocked mutants of corresponding genes. After blocking these genes, paper-disc agar diffusion assay was performed to determine as to whether bacilysin production is affected. ComQ-ComX and ComP were found to be essential for bacilysin biosynthesis. On the other hand, the disruption of srfA operon in the bacilysin producer resulted in a bacilysin-negative phenotype, thus our study verified that srfA operon functions directly in the production of bacilysin. The disruption of abrB gene in spoOA-deleted mutants suppressed the bacilysin-negative phenoptype whereas the disruption of the same gene in the wild type strain resulted in about a two-fold increase in the production of bacilysin.
