Wide-type α-hemolysin (WT-αHL) is a promising protein nanopore for nucleic acid analysis in DNA sequencing. In this work, we demonstrated three modified versions of α-HL to compare the translocation behaviour of ssDNA when they pass through the nanopore. Current signal collected from the nanopore mutants during ssDNA translocation through the protein pore provides information about the sequence and structure of the ssDNA molecule. Furthermore, the translocation velocity of ssDNA was remarkably reduced with the modifications of WT-αHL, facilitating high fidelity reads for DNA sequencing.
