Bu çalışmada tarımda zararlı otlarla mücadele amacı ile dünya çapında yaygın olarak kullanılan organoklorlu bir herbisid olan atrazinin, seçilmiş bir beyaz-çürükçül fungus suşuyla biyoyıkımı ve biyoyıkımının gerçekleştiği optimum koşullar araştırıldı. Ortamdaki atrazin miktarı HPLC analizleri ile saptandı. Atrazin biyoyıkımını gerçekleştiren en verimli suşu seçmek için beyaz çürükçül funguslardan Pleurotus sajor-caju, Phanerochaete chrysosporium, Trametes versicolor suşları denendi ve çalışma sonucunda en verimli suş olarak P.chrysosporium seçildi.Gerçekleştirilen biyoyıkım deneyleri sonucunda uygun inkübasyon zamanı 5 gün ve uygun ortam pH değeri 3.0 olarak bulundu. Ayrıca uygun inkübasyon sıcaklığı 30? C ve uygun çalkalama hızı 150 r.p.m olarak tespit edildi.Yapılan çalışmalar sonucunda lignin peroksidaz ve mangan peroksidaz enzimlerin in atrazinin biyoyıkımında rolü olmadığı tespit edildi. Araştırılan inkübasyon sıcaklığı, inkübasyon süresi ve çalkalama hızı gibi parametreler açısından üremeyle biyoyıkım arasında doğrudan bir ilişki olduğu tespit edildi. Atrazine is an organochloride herbicide widely used for controlling grassy weeds in agriculture. In this study, biodegradation of atrazine with a white-rot fungal strain and the optimum conditions of biodegradation were investigated. The amount of atrazine in the medium was determined through HPLC analysis. Among the white fungal strains, Pleurotus sajor-caju, Phanerochaete chrysosporium, Trametes versicolor were tested in order to select the most efficient strain which carries out the atrazine biodegradation. The study concluded that the most efficient strain was P.chrysosporium.The biodegradation experiments were carried out indicated that the optimum incubation time was 5 days and the optimum medium pH was 3.0. In addition, the optimum incubation temperature was determined as 30? C and the optimum agitation rate was determined as 150 r.p.m in the relevant experiments.The results of these experiments showed that the lignin peroxidase and manganese peroxidase enzymes do not have any effects on atrazin biodegradation. It was found that there is a direct relationship between growth and biodegradation when the parameters like incubation temperature, incubation time and agitation rate is taken into consideration. 51
