•A 3D printed scaffold/hydrogel composite was fabricated to provide the topological cue and compliance cue at the same time for cell guidance.•The larger pore size and bigger directed fiber number of 3D scaffold lead to a more uniform cell orientation (topotaxis).•The cells in collagen can still sense the scaffold topological cue and migrate along it (durotaxis).
