Nitro fatty acid (NO2-FA)은 불포화 지방산이 반응성 질소 종과 반응할 때 형성되는 내인성 지질 매개체로, 동물 시스템에서 낮은 나노몰 농도로 검출되지만, 염증 조건에서 수치가 크게 증가하였으며, 여러 세포 보호 및 항염증 특성을 발휘하는 것으로 밝혀졌다. 이러한 특성으로 인해 만성 염증과 관련된 다양한 병리학적 생물학적 모델에서 NO2-FA를 외인성 투여하여 치료하는 연구가 많이 진행되었지만, 염증성 치주질환의 예방 및 치료에서 NO2-FA의 효능에 대한 연구 및 보고가 아직 이루어지지 않았다. 이에 본 연구에서는 쥐 대식세포인 RAW264.7 cells에서 prevotella intermedia의 lipopolysaccharide (LPS)에 의해 활성화된 염증 반응에 대해 전형적인 NO2-FA인 nitrooleic acid이 미치는 영향을 조사하였다. 배양 상층액으로 nitric oxide (NO), inflammatory cytokines (interleukin-6, interleukin-1β) 분석을 수행하고 real-time polymerase chain reaction을 사용하여 유도성 NO 합성효소 (iNOS)의 메신저 RNA (mRNA)를 정량화하였다. Immunoblotting을 수행하여 Nuclear factor kappa-light-chain-enhancer of transcription B cells (NF-κB) p65, NF-κB p50, iNOS, inhibitory κB (IκB)-α, heme oxygenase-1 (HO-1)의 단백질 수준을 분석하였다. 또한 분비된 배아 alkaline phosphatase reporter assay을 사용하여 NF-κB 활성화를 측정하고, transcription factor assay kit를 사용하여 NF-κB subunits의 DNA 결합을 측정하였다. Nitrooleic acid은 iNOS 유래 NO와 IL-6, IL-1β의 mRNA 및 protein 발현을 억제하고 M2 표현형과 HO-1 발현을 증가시켰으며, P. intermedia LPS로 활성화된 RAW264.7 세포에서 NF-κB 활성도 억제하였다결과적으로 nitrooleic acid은 P. intermedia LPS에 의해 자극된 RAW264.7 세포에서 iNOS 유래 NO, IL-6 및 IL-1β 생성을 억제하였다. 또한 nitrooleic acid은 M1 대식세포 표현형에서 M2 대식세포 표현형으로 분극화를 촉진했으며, 이는 HO-1 유도 및 NF-κB 신호 억제를 통해 이루어졌다. 따라서 nitrooleic acid이 치주 질환 치료에서 새로운 종류의 숙주 조절제로 유용할 수 있을 것이라 사려 된다.
Nitro fatty acid (NO2-FA) is an endogenously produced lipid mediator formed when unsaturated fatty acids react with reactive nitrogen species and, although detected at low nanomolar concentrations in animal systems, has been shown to exert several cytoprotective and anti-inflammatory properties, with levels significantly increased in inflammatory conditions. Due to these properties, many studies have been conducted on the treatment with exogenously administered NO2-FA in various pathological biological models related to chronic inflammation, however there are no researches on the benefits of NO2-FA in the prevention and treatment of inflammatory periodontal disease. Accordingly, the present study investigated the effect of nitrooleic acid, a prototypic NO2-FA, on the inflammatory responses activated by lipopolysaccharide (LPS) from Prevotella intermedia in RAW264.7 cells, a murine macrophage. Nitric oxide (NO), inflammatory cytokines (interleukin-6, interleukin-1β) assays were performed with culture supernatants and real-time polymerase chain reaction was used to quantify of messenger RNA (mRNA) of inducible nitric oxide synthase (iNOS). Immunoblotting was performed to analyse protein levels of Nuclear factor kappa-light-chain-enhancer of transcription B cells (NF-κB) p65, NF-κB p50, iNOS, inhibitory κB-α (IκB-α), heme oxygenase-1 (HO-1). In addition, NF-κB activation was measured using a secreted embryonic alkaline phosphatase reporter assay and DNA binding of NF-κB subunits was measured using a transcription factor assay kit. Nitrooleic acid inhibited mRNA and protein expression of iNOS-derived NO and IL-6, IL-1β, increased M2 phenotype and HO-1 expression, and also inhibited NF-κB activity in RAW264.7 cells activated by P. intermedia LPS. It also promoted polarisation from M1 to M2 macrophage phenotype, which was achieved by HO-1 induction and inhibition of NF-κB signalling. Therefore, nitrooleic acid may be useful as a new class of host modulators in the treatment of periodontal disease.
