인체의 피부에는 다양한 미생물들이 균형을 이루면서 살고 있으며, 이들은 피부의 건강상태와 관련되어 있다. 따라서 피부 상재균을 검출하는 것은 피부 과학에서 중요한 의미를 가진다. 피부에서 가장 대표적인 상재균은 여드름균이라 불리는 Cutibacterium acnes로, 피부의 피지를 영양분으로 사용하며 대량으로 증식하면 염증을 일으키기도 한다. 이러한 피부 상재균의 신속한 검출을 위해 LAMP(loop-mediated isothermal amplification) 시스템을 개발하였다. LAMP를 수행하기 위해 Geobacillus stearothermophilus로부터 genomic DNA를 추출하여 Bst DNA polymerase large fragment 유전자를 PCR을 통해 분리하였다. 증폭된 Bst DNA polymerase 유전자를 pET-28a(+) vector에 subcloning 한 후, 최종적으로 E. coli의 BL21 (DE3)에 형질전환하여 IPTG induction을 통해 발현시켰다. 발현된 단백질은 SDS-PAGE로 분자량을 확인하였다. 배양 방법과 배양 시간 및 온도, IPTG의 농도를 조정하여 최적의 발현 조건을 확인하였다. 발현된 단백질은 Ni2+ affinity chromatography를 통해 정제한 다음, activity assay로 정확한 unit을 측정하였다. 최적의 활성을 갖는 중합효소 변이체를 찾기 위해 N-terminal에 아미노산을 추가하여 같은 방법으로 cloning 후 정제하였다. 정제된 단백질들은 activity assay를 통해 정확한 unit을 측정하여 비교하였다. 개발된 효소를 이용하여 C. acnes의 16S rRNA를 target gene으로 하는 LAMP 시스템을 구축하였고, 65 ℃에서 1시간 동안 반응하여 약 200 bp DNA 단편의 증폭에 성공하였다. 상기의 결과는 향후 피부에 상재한 미생물을 등온 조건에서 신속하게 검출할 플랫폼으로써 피부 건강을 측정하거나 피부 질환의 진단에 활용할 수 있을 것으로 기대된다.
Various microorganisms live in balance on the human skin, and they are related to the health of the skin. Therefore, detection of skin flora has important implications in dermatology. One of the most representative skin flora is Cutibacterium acnes, commonly known as acne bacteria, which utilizes sebum as a nutrient and can cause inflammation when overgrown. For rapid detection of skin flora, a Loop-Mediated Isothermal Amplification (LAMP) system was developed. To perform LAMP, genomic DNA was extracted from Geobacillus stearothermophilus and the Bst DNA polymerase large fragment gene was isolated through PCR. After subcloning the amplified Bst DNA polymerase gene into the pET-28a(+) vector, it was ultimately transformed into BL21 (DE3) of E. coli and expressed through IPTG induction. The expressed protein was confirmed using SDS-PAGE to determine its molecular weight. Optimal expression conditions were established by adjusting culture methods, duration, temperature and IPTG concentration. The expressed protein was purified using Ni2+ affinity chromatography, followed by measuring the precise unit through an activity assay. To find polymerase variants with optimal activity, amino acids were added to the N-terminal and then cloned and purified in the same method. After purification, the proteins were compared by measuring the precise units through an activity assay. Utilizing the developed enzyme, a LAMP system targeting the 16S rRNA of C. acnes was constructed, successfully amplifying approximately 200 bp DNA fragments during a reaction at 65 °C for 1 hour. The above results are expected to be used to measure skin health or diagnose skin diseases as a platform to quickly detect skin flora under isothermal conditions in the future.
