무(Raphanus sativus L.)는 십자화과에 속하는 뿌리채소로, 전 세계적으로 널리 재배되고 소비되고 있다. 무는 항산화, 항암, 항균 및 항당뇨 등의 다양한 생리활성을 발휘하고, 플라보노이드류, 테르펜류, 글루코시네이트류 등 다양한 성분을 함유하고 있다. 무는 4대 채소로 널리 이용되면서 다양한 품종이 개발되고 있으며, 유색무 또한 재배되고 있다. 특히, 안토시아닌류를 함유하고 있는 자색무는 건강 기능성이 우수하다고 인식되고 있다. 그러나, 자색무는 안토시아닌류를 비롯한 성분학적 특성에 대한 객관적 근거가 미흡한 실정이다. 따라서 본 연구에서는 백색무와 자색무의 항산화 활성과 LC-MS 기반 대사체 분석을 통해 성분학적 특성을 파악하고자 하였다. 또한, 자색무로부터 안토시아닌류을 분리 및 정제한 다음, 그 정확한 구조를 밝히고자 하였다. 백색무 1종(무병장수)과 자색무 2종(스위트베이비, 정운무)의 무청과 무를 대상으로 항산화 활성 및 대사체 분석을 실시하였다. 그 결과, 자색무의 무청과 무는 백색무에 비하여 ABTS+ 라디칼 소거 활성과 철 환원력이 유의적으로 높았으며, 무는 라디칼 소거 활성이, 무청은 철 환원력이 우수함을 알 수 있었다. 자색무의 무청과 무는 백색무에 비해 총 페놀성 화합물 함량과 총 플라보노이드 함량이 높았으며, 무에 비해 무청에 더 높은 함량이었다. 또한 자색무의 과육과 껍질을 대상으로 총 안토시아닌 함량을 비교하였고, 과육보다는 껍질에 더 높은 함량이었다. 이어 백색무와 자색무의 무청과 무를 대상으로 LC-MS 대사체 및 다변량 통계분석을 실시하였다. 무청의 LC-MS (positive와 negative) chromato gram에서 19종의 주요 피크들이 관찰되었으며, 그 주요 화합물은 대부분 querce tin과 kaempferol을 포함한 flavonoid 배당체들로 동정되었다. PCA score plot 에서는 백색무 1종과 자색무 2종의 무청 시료들 간에 확연하게 구별되었고, 정운무에는 quercetin 및 kaempferol 배당체가, 스위트베이비에는 quercetin ruti noside가, 그리고 백색무는 apigenin 배당체가 상대적으로 높은 함량인 것으로 나타났다. 무의 경우 LC-MS (positive와 negative) chromatogram에서 27종의 주요 피크들이 관찰되었으며, 이 중 3종의 quercetin 배당체와 22종의 cyanidin 류가 동정되었다. 무 또한 자색무와 백색무 시료들 간에 확연하게 구별이 되었으며, 자색무 2종에는 cyanidin 배당체들이, 백색무에는 quercetin glucuroni de가 주요 화합물인 것으로 나타났다. 자색무인 정운무와 스위트베이비에 함유된 안토시아닌은 대부분 cyanidin류인 것으로 확인되었다. 한편, 자색무에 함유된 안토시아닌류을 분리•정제하고 그 정확한 구조를 밝히고자, 자색무 MeOH 추출물의 water-saturated n-butanol (BuOH)층을 대상으로 용출용매 조건를 달리하여 다수의 ODS column chromatography를 실시하여 화합물 1+2를 분리하였다. 분리된 화합물들은 HR-ESI-MS 및 NMR 분석을 통해 cyanidin 3- O-[2-(O-6-p-coumaroylglucosyl)-6-O-feruloyl] glucoside 5-O-(4-glucosyl -6-O-malonyl)glucoside (1)와 cyanidin 3-O-[2-(O-6-p-coumaroylglucosyl) -6-O-sinapoyl]glucoside 5-O-(4-glucosyl-6-O-malonyl) glucoside (2)로 구조해석되었다. 현재 자색무에 함유된 다양한 안토시아닌류를 추가적으로 구명 중에 있다. 이러한 안토시아닌류를 기반으로 한 자색무의 성분 데이터베이스가 구축되어진다면, 재배, 가공, 생산, 저장 및 유통에 이르기까지 품질관리의 지표성분으로써의 활용이 기대된다.
Radish (Raphanus sativus L.), cruciferous family, is a root vegetable that is grown and consumed worldwide. Radish has been reported to exert various biological effects, including antioxidant, anticancer, antimicrobial, and antidiabetic activities. Radish contains several health beneficial compounds, such as flavonoids, terpenes, and glucosinolates. Purple radish contains various health-promoting acylated anthocyanins that can effectively scavenge free radicals, suppress inflammation, and prevent cancer, diabetes, and cardiovascular diseases. Recently, cyanidin glycosides having organic acids and phenylpropanoids were identified in purple radish using a LC-MS-based metabolomic analysis. However, studies on structural determination of anthocyanins in purple radish are limited. In this study, the antioxidant activity and several bioactive compound characteristics of white and purple radish were compared and anthocyanins from purple radish were also purified and structural-determined. Purple radish showed significantly higher radical scavenging activity and iron reducing capacity than white radish. Purple radish had higher total phenolics and flavonoid content compared to white radish. Total anthocyanin content of purple radish had higher in the peel than in the flesh. Purple radish root and leaves were analyzed by LC-MS-based metabolomic analysis. LC-MS results indicated that quercetin and kaempferol glycosides were as the main compounds in the purple radish roots. Additionally, two anthocyanins from water-saturated n-butanol (BuOH) layer of purple radish methanolic extract was purified by ODS column chromatography. Two anthocyanins were elucidated as cyanidin 3-O-[2-(O-6-p-coumaroylglucosyl)-6-O-feruloyl] glucoside 5-O-(4-glucosyl-6-O-malonyl)glucoside and cyanidin 3-O-[2-(O- 6-p-coumaroylglucosyl)-6-O-sinapoyl]glucoside 5-O-(4-glucosyl-6-O- malonyl)glucoside, based on MS and NMR analyses.
