질산염은 다양한 출처로부터 지하 표면으로 유입되었습니다. 토양으로 유입된 질산염은 지하환경의 지하수로 유입되어 지하환경을 교란시키며, 오염물질로 생성되어 인체에 직·간접적으로 영향을 미쳐 위험을 발생시킵니다. 그러나 국내 기술은 지하환경 관리에 소홀하여, 유입경로인 토양 정화에 초점을 맞춰 기술 개발을 진행해 왔다. 이들 대부분은 토양을 직접 처리하는 기술로 정화 비용이 많이 든다. 오염지역의 부지는 토양/지반환경이 정화될 때까지 이용이 불가능하다는 한계가 있다. 도시지역/산업시설의 오염은 상대적으로 심각할 수 있으나 그 특성상 부지 활용도를 저하시키지 않는 기술이 필요하다. 직접처리 기술은 높은 환경부하를 발생시켜 토양의 질을 저하시키는 단점이 있다. 이러한 이유로 자연 저감 기술 개발이 시급하며, 느린 정화 속도를 개선할 수 있는 촉진 기술 개발이 필요하다.본 연구에서는 질산염 제거를 위한 자연 감쇠 가능성을 조사하였다. 자연저감 촉진을 평가하기 위해 실내조사를 통해 최적의 탄소원과 그 농도를 파악하였고, 실내 연구 결과를 바탕으로 MbT-Tool을 이용한 최적의 탄소 주입 농도를 결정하였다. 결정된 탄소원 농도를 통해 충적대수층과 기반대수층에 탄소원 주입 전후의 탈질률을 평가하여 자연저감 촉진을 검증하였다. 연구 결과, Paracoccus denitrificans의 성장촉진 스펙트럼 C/N 비율은 C:N = 2.1:1인 것으로 나타났다. 이때 미생물 증식촉진에는 아세테이트가 가장 많이 관여한다는 사실이 기초연구를 통해 확인되었다. 이러한 결과를 바탕으로 C/N 비율을 동일하게 유지하여 포장지하수에 존재하는 미생물의 생장을 촉진시키는 포장투입조건도 평가하여 탈질촉진 가능성을 확인하였다. 암반층과 충적층도 동일한 탄소원으로부터 동일한 비율과 성장을 보였다. 탈질촉진 후 암반층에서는 0.30 mg N/L day에서 5.10 mg N/L day로 가속되는 것을 확인하였다. 탈질촉진 후 발생하는 미생물의 군집분석을 통해 탈질미생물 개체수의 증가를 확인하였고, 탄소원 주입을 통한 질산성질소에 의한 탈질촉진을 확인하여 암반층 내에서 acetate를 이용한 탈질 촉진을 검증하였다. 충적층 연구는 현장 실증을 통해 탄소원 주입에 의한 생물학적 탈질화의 질산염 감소 효율을 조사하였다. 현장 테스트는 현장 처리의 질산염 감소 효율을 평가하기 위해 22일 동안 아세테이트를 탄소원으로 사용하여 연구를 진행하였다. 탄소원 주입 전후에 next-generation sequencing을 이용한 지구화학적 매개변수 및 미생물군집 분석을 수행하였다. 12시간의 반응 시간 후 질산염 농도는 PC-2에서 31.6에서 4.2 mg-N/L로 감소한 후 3.9 mg-N/L에서 안정적으로 유지되는 것을 확인하였다. 아세테이트 주입 시 질산염 감소율은 29.0 mg-N/L/day로 나타났으며, Aquabacterium commune, pseudomonas Brassicacearum, declomonas denitrificans 및 Massilia FAOS는 acetate 주입 후 우점종으로 확인되었다. 예측 대사 경로 분석에 따르면 아세테이트 주입 중 질산염 환원 대사는 탈질화 및 암모늄으로의 동화성 질산염 환원으로 나타났고, 질산염으로 오염된 지하수의 평가된 위험지수는 아세테이트 주입 후 크게 감소된 것을 확인하였다(어린이의 경우 비발암성 위험도가 1.176에서 0.134로 감소). 암반층 연구와 다르게 N2O 분석을 통한 탈질을 검증하였으며, N2O 농도가 탈질촉진 후 약 1.9배 증가한 결과를 확인할 수 있었다 (탈질 촉진 전 352 μg L-1 -> 탈질 촉진 후 639.6 μg L-1).
Nitrate that flows into the soil in turn flows into groundwater present in the underground environment, thereby disturbing it. This leads to pollutant generation that directly or indirectly affects the human body, posing health risks. However, domestic technologies have been negligent in managing the underground environment, and technology development has been conducted with a focus on soil purification, which is the inflow path. Most of these approaches involve direct soil treatment technologies that result in high purification costs. One limitation is that the site in the contaminated area cannot be used until the soil/ground environment is cleaned. Pollution in urban areas/industrial facilities may relatively be serious. Moreover, considering their characteristics, a technology that does not reduce site utilization is necessary. However, direct treatment technologies reduce the soil quality by generating a high environmental load. Therefore, the development of a natural abatement technology is urgently needed, along with a promotion technology, which can improve the slow purification speed.This study investigates the potential of natural attenuation for nitrate removal. Through indoor research, the optimal carbon source and its concentration are identified to evaluate the promotion of natural reduction. The optimal concentration of carbon injection is determined using the MbT-Tool based on the indoor study results. The natural reduction acceleration is verified by evaluating the denitrification rate before and after carbon source injection into the alluvial and bedrock aquifers through the determined carbon source concentration.The results show that the growth promotion spectrum C/N ratio of Paracoccus denitrificans is 2.1:1. Basic research confirms that during this time, acetate is most involved in stimulating microbial growth. Considering these outcomes, the conditions for injecting substances into the field are assessed to enhance the development of microorganisms in the field's groundwater through the preservation of the C/N ratio, thereby confirming the possibility of stimulating denitrification. Furthermore, the rock and alluvial layers have the same ratio and growth from the same carbon source.After denitrification stimulation, the reduction rate of nitrate is found to accelerate from 2.6 to 29.0 mg N /L day in the alluvial layer and 0.30 to 5.10 mg N /L day in the rock layer. Moreover, an increase in the population of denitrifying microorganisms is found through a community analysis of the microorganisms generated after the stimulation denitrification. Additionally, the promotion of denitrification by nitrate nitrogen through carbon source injection is verified.
