Atopic Dermatitis (AD) and Psoriasis (PSO) are prevalent chronic inflammatory skindiseases associated with pruritus. To explore the pathological disparities between AD and PSO, we conducted a comparative study using publicly available single-cell transcriptome data. We identified an AD-specific Fibroblast subtype, COL6A5+ fibroblasts, and PSO-specific Inflammatory Keratinocytes subtype, both playcrucial roles in skin abnormalities and neuronal regulation and projection guidance. Moreover, the inference of unique interactions between disease-specific subtypes and skin neurons implicated their potential role in determining pruritus in AD and PSO respectively. These findings illuminate the pathological distinctions underlying pruritus in AD and PSO presenting promising avenues for further study on pruritus.
