Radishes (Raphanus sativus L.) are a globally cultivated and widely consumed vegetable, characterized by a diverse array of shapes, sizes, and colors. Radish is a root vegetable classified within the Brassicaceae family, a botanical group that also encompasses prominent crops such as cabbage, broccoli, and mustard. Radish is a vital crop due to its edible taproot, making nutritional growth a critical factor. Ealy flowering directly affects the marketability of radishes, necessitating the development of radish cultivars with delayed flowering. Therefore, this research aimed to create late-flowering radish lineages. In this study, three homologs of FLC (FLOWERING LOCUS C) were initially identified through transcriptome analysis in radish. These homologs were subsequently transformed into Arabidopsis thaliana for phenotype confirmation. Notably, all three FLC homologs played an important role in inhibiting the transition to flowering. This study aim is to gain molecular insights into the mechanism by which FLC genes are suppressed in radishes. As a result, RsFLC1 required the RY-motif for inhibiting flowering. Brassicaceae family, are known for their high source of glucosinolate content. Glucosinolates (GSLs) are a class of sulfur-containing compounds found in various cruciferous vegetables. These compounds are primarily responsible for the bitter and pungent tastes that can potentially compromise the flavor of vegetables. However, they exhibit potent anticancer properties. Therefore, this study aimed to reveal major genes responsible for GSLs biosynthesis in radish. It might provide the breeding strategy enhancing marketability by adjusting the GSLs content in radish.By studying thirteen radish lineages with diverse phenotypic variations available in the market, it was confirmed that RsGRS1 is a crucial gene responsible for the glucosinolate biosynthesis, particularly for the glucoraphasatin (GRH) in radishes. Overexpression and knock-down of RsGRS1 using RNA interference (RNAi) strategy were employed to modulate the endogenous contents of GSLs in radish. In summary, this study provides insights on the molecular mechanism underlying vernalization-mediated floral transition in radish and suggests useful molecular breeding approaches to modulate GSLs content, which might enhance market value of radish plant.
무(Raphanus sativus L.)는 전 세계적으로 재배되며 다양한 모양, 크기 및 색상을 가지고 있다. 무는 Brassicaceae 과에 속하는 작물로 주로 뿌리를 식용하는 대표적인 작물로써 영양생장이 중요 하다. 여러 환경적 요인에 의해 발생하는 조기 개화는 무의 상품성에 직접적으로 부정적인 영향을 줌으로 개화시기가 늦은 무 품종 개발이 요구 되고 있다. 본 연구에서는 이러한 요구에 따라 만추대성무 계통을 만들고자 하였다.먼저 춘화처리 된 무에서 실시한 전사체 분석 결과 무에서 3개의 FLC (FLOWERING LOCUS C) homologs를 찾아냈다. 이렇게 찾아진 무의 FLC homolog들은 애기장대에 형질전환하여 표현형 검정을 실시하였다. 그 결과 3개의 FLC homolog 모두 개화로의 전환을 억제하는 데 중요한 역할을 하는 것으로 확인되었다. 그렇다면 무의 FLCs 유전자가 어떠한 분자적인 기전으로 춘화기간 동안 발현이 억제되는지에 대한 메커니즘을 밝히고 자 시도하였다. 그 결과 무에서 찾은 FLC homolog 중 RsFLC1은 해당 유전자가 억제될 때 RY-모티프가 필요하다는 것을 본 연구내용을 통해 확인하였으며, 이에 따라 CRISPR-Cas9 기술을 활용하여 RsFLC1의 RY-motif를 제거하여 만추대성 무를 만들고자 하였다.한편 십자화과의 작물들은 높은 글루코시놀레이트를 함유하고 있다. 글루코시놀레이트는 십자화과 작물에서 발견되는 황 함유 화합물의 일종으로 이러한 화합물은 주로 쓴맛 및 매운맛을 유발하여 채소의 풍미를 떨어뜨리기도 하지만 강력한 항암 효과를 지니고 있다. 따라서 본 연구에서 는 무의 글루코시놀레이트 함량이 조절된 무 계통을 만들고자 하였다. 이를 위해 시중에서 판매되고 있는 13개의 다양한 무 계통을 qRT-PCR과 GSLs 함량을 분석함으로써 RsGRS1이 무에서 GSLs합성에 중요한 역할을 한다는 것을 규명하였다. 그리고 RsGRS1을 무에서 과발현시키거나 발현을 억제함으로써 무 계통 중에서 항암 특성이 뛰어나거나 풍미가 향상된 무를 제시하였다. 따라서 본 연구는 무의 늦은 개화 특성을 보이는 만추대성 계통과 글루코시놀레이트 함량이 조절된 무 계통을 제시함으로써 그 시장 가치를 향상시키고 앞으로의 무 육종 연구에 많은 기여를 할 것으로 사료된다.
