The purpose of this experiment is to improve the mechanical properties of hydrogel. This improvement is to be achieved by the photo-crosslinking of gelatin and alginate, which have excellent biocompatibility. Additionally, the experiment also sought to synthesize a hydrogel with excellent antibacterial activity, this to be achieved by loading catechin, a natural product with excellent antibacterial properties. Bacterial infection has a high mortality rate worldwide. Antimicrobials play the most important role in fighting infectious diseases, and their antibacterial activity can kill bacteria locally or limit their growth. However, the emergence of bacterial resistance, itself caused by excessive use and abuse of antimicrobials, bacterial resistance is an increasing problem.Catechin, a natural product derived from green tea, possesses antibacterial, anticancer, and anti-inflammatory activity. In particular, one of the characteristics of it being a natural product is that its inherent antibacterial activity can be sustained over the long term due to bacterial resistance to it being remarkably low.Hydrogels are widely used in drug delivery systems. This is due to their excellent biocompatibility, the ability to control the dissolution rate and drug release which is derived from their physical properties. Hydrogels prepared using natural polymers, such as alginate or gelatin, are excellent with regards biocompatibility and biodegradability. This makes them extremely useful when skin contact is required and for delivering drugs locally. Synthesis of hydrogels using a photo-crosslinking agent is a method that has recently been widely adopted due to the fact that it can enhance mechanical properties in a short time, as compared to other crosslinking methods.We synthesized alginate and gelatin using methacrylic anhydride and then synthesized methacrylate alginate (AlgMA) and methacrylate gelatin (GelMA) through photocrosslinking. In addition, a hydrogel was prepared using the photoinitiator 2-Hyroxy-4'-(2-hydroxyethoxy)-2-methylpropiophenone (I2959) and UV light (365 nm). The prepared GelMA hydrogel showed both a low disintegration rate and a high expansion rate. Furthermore, it was confirmed that it had uniform pores, this being determined by means of SEM imaging. In addition, when the Gel/Alg MA (GAMA) hydrogel was synthesized and its mechanical properties compared to that of GelMA hydrogel, it was confirmed that they had similar physical properties. The drug release from the GAMA hydrogel, loaded with catechins, was confirmed by means of absorbance measurement, while a catechin toxicity test (at each concentration) was performed on Human Dermal Fibroblast (HDF). The cell viability of 0.005 and 0.01 M catechin displayed a high survival rate of about 90%, and it was confirmed, by means of a Live/Dead assay, that the cells grew well when the HDF was cultured on the GAMA hydrogel. Finally, the antibacterial activity of the GAMA hydrogel, loaded with catechin, was tested using a disk diffusion assay. This assay confirmed that the zone of inhibition by use of catechin occurred in 4 types of Gram-positive bacteria and 2 types of Gram-negative bacteria. To confirm that the bacteria were indeed killed by catechin, a Live/Dead assay was performed using Staphylococcus aureus, a representative gram-positive bacterium, and Escherichia coli, a gram-negative bacterium. It was found that the dead distribution was high in catechin-treated bacteria. Given this, a gelatin/alginate MA hydrogel with low toxicity and excellent mechanical properties was prepared, and a hydrogel with sustained drug release and high antibacterial activity was developed.
이 실험 목적은 생체적합성이 뛰어난 젤라틴과 알지네이트의 광가교 결합을 통한 하이드로겔의 기계적 물성 향상과 항균성이 뛰어난 천연물인 카테킨을 로딩하여 항균 활성이 뛰어난 하이드로겔을 합성하는 것이다. 박테리아를 통한 감염은 전 세계적으로 높은 사망률을 기록하고 있다. 항균제는 전염병과 싸우는데 가장 중요한 역할을 하며, 항균 활성은 박테리아를 국소적으로 죽이거나 성장을 제한할 수 있다. 하지만 항균제의 과도한 사용과 오남용으로 인해 생기는 박테리아 내성의 출현 이후 지속적으로 내성이 강해지고 있다. 카테킨은 녹차에서 나오는 천연물로써 항균성, 항암성 및 항염증 활성 효과를 가지고 있다. 특히 천연물이라는 특성은 박테리아 내성이 현저히 낮기 때문에 항균 활성을 오래 유지할 수 있다. 하이드로겔은 우수한 생체 적합성, 물리적 특성의 조절로 인한 분해 속도 및 약물 방출 제어가 가능하여 약물 전달 시스템에서 널리 사용되고 있다. 특히 알지네이트 혹은 젤라틴과 같은 천연 고분자를 이용하여 제조된 하이드로겔은 생체 적합성 및 생분해성 능력이 뛰어나 피부에 접촉되어 국소적으로 약물을 전달할 때 유용하게 사용된다. 광가교제를 이용한 하이드로겔의 합성은 다른 가교 결합 방식에 비해 기계적 물성을 짧은 시간 내에 강화시켜줄 수 있기 때문에 최근 많이 채택되고 있는 방식이다. 우리는 methacrylic anhydride를 이용하여 알지네이트와 젤라틴을 합성하고 광가교결합을 통한 methacrylate alginate (AlgMA)와 methacrylate gelatin (GelMA)을 합성하였다. 또한 광개시제인 2-Hyroxy-4’-(2-hydroxyethoxy)-2-methylpropiophenone (I2959)와 UV light (365 nm)를 이용하여 하이드로겔을 제작하였다. 제조된 GelMA 하이드로겔은 낮은 분해율과 높은 팽창율을 보여주었으며, SEM 이미지를 통해 균일한 기공을 가지고 있다는 것을 확인하였다. 또한 Gel/Alg MA (GAMA) hydrogel을 합성하여 GelMA 하이드로겔과의 기계적 물성을 비교하였을 때 비슷한 물성을 가지고 있다는 것을 확인하였다. 카테킨이 로딩된 GAMA 하이드로겔은 흡광도 측정을 통해 약물 방출을 확인하였으며, Human Dermal Fibroblast (HDF)에 대한 농도별 카테킨 독성 테스트를 진행하였다. 0.005, 0.01 M 카테킨의 세포 생존율은 약 90%로 높은 생존율을 보여주었으며, GAMA hydrogel 위에 HDF를 배양했을 때 세포가 잘 성장하는 것을 Live/Dead assay를 통해 확인하였다. 마지막으로 disk diffusion assay를 통해 카테킨이 로딩된 GAMA 하이드로겔의 항균 활성을 테스트 하였으며, 4종의 그람 양성균, 2종의 그람 음성균에서 카테킨에 의해 zone of inhibition이 발생하는 것을 확인하였다. 카테킨에 의해 박테리아가 죽는 것을 확인하기 위해 대표적인 그람양성균인 포도상구균과 그람음성균인 대장균을 이용하여 Live/Dead assay를 진행하였는데, 카테킨 처리된 박테리아에서 Dead 분포가 높게 나타났다. 이로써 낮은 독성을 가지고 기계적 물성이 뛰어난 Gelatin/alginate MA 하이드로겔을 제조하였으며, 지속적인 약물 방출과 높은 항균 활성 성능을 가진 하이드로겔을 개발하였다.
