Volatile organic solvents are widely used in the extraction and separation processes despite their potential to pollute the environment and undermine human health. Therefore, the exploration of green and versatile solvents with safe compositions has been in urgent demand. Herein, eutectic mixture-based solvents were investigated as potential green solvents for analytical chemistry applications. First, thymol-based hydrophobic deep eutectic solvents were in situ formed and applied in a simple and ecofriendly sample preparation method for antibiotics analysis in surface water. The developed method based on liquid-liquid microextraction coupled to liquid chromatography is simple, quick, easy to perform, reliable and requires no toxic or volatile organic solvents for the sample preparation procedure. Secondly, based on the thermodynamics modeling and machine learning algorithms, various carvone-based mixtures were formed and found to have polar protic to dipolar aprotic properties, which may allow them to substitute for several types of conventional solvents with similar properties. Their further application to extraction of model dyes and laver samples proposes that carvone and its mixtures could broaden the pool of hydrophobic green solvents with tunable and biodegradable properties to substitute conventional solvents for green separation. Thirdly, an effective extraction and purification of highly valued fluorescent protein, phycoerythrin using eutectic mixtures was investigated. Based on quantum chemistry modeling and microscopic visualization, proline-based mixtures were found to have strong effects on the disruption of the exopolysaccharide layer and extraction of phycoerythrin. Further aqueous two-phase system-based procedure allowed simple isolation of phycoerythrin with high purity. This study demonstrated that eutectic mixtures have promising potential as green and effective solvents in protein extraction and purification field. Collectively, current studies provide that eutectic mixture-based solvents are green and alternative solvents to hazardous solvents in various application forms of analytical chemistry.
휘발성 유기용매는 추출과 분리 공정 등 여러 분야에서 널리 사용되고 있으나 환경오염과 인체 유해성 등의 문제로 인해 최근 안전하고 친환경적인 신규 용매 개발이 요구되고 있다. 본 연구에서는, 공융혼합물(eutectic mixture)을 분석화학 분야에서 사용할 수 있는 친환경 용매로서 활용하고자 다양한 기초 및 응용 연구를 수행하였다. 첫 번째로, thymol 기반 소수성 eutectic mixture를 in situ 합성하여 지표수 내 오염물질인 항생제 분석을 위한 쉽고 친환경적인 시료 전처리법을 개발하였다. 이 방법은 휘발성 유기용매를 대신하여 친환경 eutectic mixture을 이용한 액상미량추출법 기반 시료 전처리법을 통해 용이하고 신뢰할 수 있는 분석법 개발이 가능함을 증명했다. 두 번째로, 열역학 모델링과 기계학습 알고리즘을 통해 여러 종류의 carvone 기반 eutectic mixture를 디자인하였고, 이들이 극성 프로톤 용매부터 쌍극자 비프로톤 용매까지 폭넓은 특성을 갖기 때문에 이들과 유사한 특성을 가진 기존의 여러 용매를 대체할 수 있음을 확인하였다. 이들 혼합물들은 생분해성이 우수하였으며, 실제로 소수성 및 친수성 모델 염료 분리에 효과적이었고, 식용 김으로부터 천연의 소수성 색소 추출에 효율성과 선택성이 뛰어남을 확인하였다. 본 연구를 통해 고효율 추출 용매로 활용 가능한 새로운 소수성 친환경 용매를 제안하였다. 세 번째로, proline 기반 eutectic mixture를 개발하여 홍조류에 있는 고부가가치의 형광 단백질인 phycoerythrin의 효율적인 추출과 정제가 가능함을 입증하였다. 열역학 모델링과 현미경 및 분광법 분석 등을 통해서 eutectic mixture가 Porphyridium sp. 미세조류의 다당류 층을 효율적으로 파괴하여 phycoerythrin을 고효율로 추출할 수 있음을 확인하였다. 또한 aqueous two-phase system을 이용한 추가 정제과정을 통해 고순도 및 안정적인 상태로 phycoerythrin을 정제할 수 있었다. 이로써 eutectic mixture를 친환경적이고 경제적인 단백질 추출 및 정제에 이용 가능함을 확인했다. 종합적으로, 본 연구는 기존 용매의 친환경 대체재로서 eutectic mixture이 다양한 분석 화학 분야에서 활용 가능함을 제안하고자 한다.
