To prevent the potential harmful effect of DNA damage, cells have evolved a variety of pathways to respond to DNA damage which are collectively termed DNA damage response (DDR). However, cells in mitosis cannot properly repair DNA as their DDR mechanism is suppressed. On the other hand, oocytes, the cell which are always in the middle of meiosis because of cell cycle arrest, can repair DNA damage according to previous studies. Although the MI (Meiosis I) stage is a must-go through stage for a progression to the MII stage for GV oocytes and is indispensable in oocyte maturation and right chromosome segregation, it is veiled if MI oocytes can repair DSBs sites and which pathway dominantly works. Here I show that MI oocytes have the ability to repair damaged DNA accompanied by recruitment of CIP2A-p-MDC1-p-TOPBP1 complex to chromosomes. DNA double-strand break (DSB) induction at meiosis I impairs spindle organization and pole integrity, which is partially recovered during progression through meiosis I. Similar to mitotic cells, oocytes recruit MDC1 and TOPBP1 to chromosomes in response to DNA damage. Also, BRCA1 is recruited to DSB sites unlike mitosis and HR repair pathway is more utilized to overcome MI arrest caused by DNA damage. I demonstrate that CIP2A and p-MDC1 interacts on spindle pole and chromosome while CIP2A and p-TopBP1 only interacts on chromosome in DDR with PLA (Proximity Ligation Assay). Therefore, results suggest that CIP2A interacts with p-MDC1 and p-TopBP1 on chromosome in response to DNA damage in MI oocytes.
DNA 손상 반응 (DDR) 이란 세포가 노화 또는 방사선, 자외선, 화학물 노출 같은 외부 자극에 의해 DNA가 손상되는 것에 대응하는 능력이다. 포유류 난자에서 DDR은 특히 중요한데, 그 이유는 만약 이러한 손상에 적절히 반응하지 못할 시 염색체 비정상을 유발하여 난임, 발생능 저하, 유전병이라는 결과를 불러올 수 있기 때문이다. 그러나 이러한 중요성에도 불구하고 MI난자에서 DDR의 많은 부분을 파악하지 못한 상태이다. 또한 DDR 만큼이나 양극성 방추사 형성도 유전체 보존에 중요한 역할을 하는데, 체세포와 난자에서 그 형성 과정이 매우 다르다. 체세포에서는 양극이 중심체에 의해 형성이 되는데, 난자에서는 중심체가 없이 양극이 형성된다. 이를 acentriolar microtubule organizing centers (aMTOCs)이라 하며, 중요성과 특이점에도 불구하고, 이것의 요소와 작동 원리는 많은 부분 밝혀지지 않았다. 최근 연구에서, 체세포에서는 PP2A의 내성 억제제로 알려진 CIP2A가 난자에서는 새로운 aMTOC의 요소이고 CEP192와 함께 Plk1과 Aurora A를 조절함이 발견되었다. 그런데 이 연구에서 aMTOC의 요소로 알려진 CIP2A가 양극이 아닌 염색체에도 존재함을 발견하였고, 본 연구에서는 CIP2A가 DNA 손상 반응에서 p-MDC1과 p-TopBP1과 함께 어떤 역할을 하는지에 대해 조사하였다. 먼저 MI난자에 Etoposide를 처리하여 이중 가닥 절단 (DSBs)을 유도한 결과, spindle이 짧아지는 것을 확인하였다. 또한 DNA 회복 시간을 주며 배양했을 시, 다시 spindle이 길어지는 것을 확인하였다. 이를 기반으로 MI 난자에서 DNA 회복이 되는 지를 확인하기 위해, 각각 HR 저해제와 NHEJ 저해제를 처리하며 DNA 회복을 시켰고, HR 저해 시 MI arrest가 일어남을 확인하였다. 또, 크로모솜 스프레딩과 TUNEL 에세이를 한 결과, BRCA1이 염색체로 모집되었고 TUNEL 시그널이 감소하였다. DNA 회복에 관여하는 단백질인 p-MDC1과 p-TopBP1이 spindle pole에 위치하다가, 난자에 DNA 손상을 가했을 시, 염색체로 그 위치를 옮기는 양상을 확인하였다. aMTOC의 요소인 CIP2A도 같은 양상을 띄는 것을 확하였고, PLA를 통해 CIP2A와 함께 p-MDC1과 p-TopBP1은 상호작용하며 그 위치를 옮긴다는 것을 알 수 있었다. 이러한 결과를 통해 CIP2A가 p-MDC, p-TopBP1과 복합체를 형성하며 MI 의 DNA 손상 반응에서 중요한 역할을 하고있다는 사실을 확인하였다.
