메티실린 내성 황색포도상구균 (MRSA) 감염은 항생제 내성 및 생체막 형성 때문에 근절하기 어렵다. 새로운 항균제와 대체요법이 시급하다. 본 연구에서는 Sanguisorba officialis L. 에서 분리된 Sguisorbigenin (SGB)의 항 MRSA 활성을 발견하고 SGB와 기존 항생제의 상승효과를 평가했다. 육수 미세희석 측정을 사용하여 항균 활성을 측정하기 위해 총 8개의 기준 균주와 임상분리균주를 사용되었습니다. SGB의 최소 억제 농도 값은 12.5-50 μg/mL 이었다. MRSA에 대한 항생제와 결합된 SGB의 상승 효과를 결정하기 위해 체커보드 희석 시험과 시간-죽이기 곡선 측정이 사용되었다. SGB는 항생제와 상당한 시너지를 보였으며 항생제의 최소 억제 농도를 2–16배 낮췄다. 시너지 메커니즘을 평가하기 위해 웨스턴블랏, 정량적 RT-PCR, 바이오필름 억제 앗세이 크리스탈 바이올렛 흡수, 투과전자현미경 검사를 수행하였다. 결과는 SGB로 치료했을 때 페니실린 결합단백질2a의 수준과 mecA 유전자의 발현이 유의미하게 감소했음을 보여주었다. 또한,SGB는 생체막 형성을 억제하고 MRSA의 세포막 투과성을 변화시킬 수 있다. 또한 SGB는 상당히 낮은 세포독성과 용혈성을 나타내는 것으로 밝혀졌다. SGB의우수성이 발견됨에 따라 SGB는 결합치료와 바이오필름을 대상으로 하는 식물 유래 항균제로 사용될 수 있는 항생제 보조제일 수 있다.
Staphylococcus aureus resistance to antibiotics has increased dramatically as a result of the overuse of antibiotics. It is crucial to develop novel antibacterial agents to alleviate the pressures of multi-drug resistance. In this research, we identified that sanguisorbigenin (SGB) isolated from Sanguisorba officinalis L. had potent antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA) and demonstrated a synergistic effect with conventional antibiotics. The checkerboard dilution assay showed that the range of minimum inhibitory concentration (MIC) values for SGB was 12.5 to 50 µg/mL, and the combination treatment decreased the MIC value of the antibiotic by 2 to 16-fold. The synergy mechanism was investigated using western blot and qRT-PCR. Data suggested that SGB could down-regulated the resistance gene mecA and decreased the expression of PBP2a in MRSA. Moreover, SGB inhibited biofilm formation and down-regulated the transcription of virulence-related genes. In addition, SGB showed fairly low cytotoxicity and hemolytic activity, and no obvious drug resistance. The study suggested that SGB might be a potential plant-derived antibacterial agent targeting biofilm and an antibiotic adjuvant.
