인간 지방유래 줄기세포는 특유의 치료능과 분화능으로 인해 재생 의료에 활용되어져 왔다. 하지만 세포를 그대로 사용하기에는 낮은 생존율과 제한된 이식 조건 등이 문제가 되어왔다. 이러한 문제를 극복하기 위한 방법 중 하나로 photobiomodulation (PBM)이 활용되곤 한다. 본 연구에서는 녹색 파장대의 organic light emitting diode (OLED) 빛을 조사한 인간 지방유래 줄기세포의 치유 효능과 치료 효과 향상에 대해 조사하였다. 조직 재생 능력 확인을 위하여 OLED 빛을 조사한 인간 지방유래 줄기세포의 증식과 이동, 부착과 관련된 유전자와 단백질 발현을 여러 가지 시험을 통해 분석하였고, in vivo 치료 효과를 분석하기 위하여 창상 모델을 이용하였다. 그 결과로, 빛을 조사한 세포의 증식과 이동, 부착, 성장 인자 발현 모두 증가했을 뿐만 아니라 빛을 조사한 인간 지방유래 줄기세포를 사용하였을 때 빛을 조사하지 않은 세포를 사용했을 때보다 더 나은 창상 치료 효과를 보여주었다. 또한 본 연구에서 사용한 특정 파장대의 빛이 인간 지방유래 줄기세포를 자극하는 기전을 확인하였다. 결과적으로, 녹색 OLED 빛 조사는 인간 지방유래 줄기세포의 치유 효능을 향상시켰으며 이를 활용한 치료 방법 발전의 여지를 남겨두었다.
Human adipose-derived stem cells (hASCs) have been used in regenerative medicine due to their unique therapeutic and differentiation capabilities. However, there are problems such as low survival rates and limited transplantation conditions to use hASCs as they are. Photobiomodulation (PBM) is often used as one of the ways to overcome these problems. This study investigated therapeutic efficacy and preclinical effects of hASCs improved by PBM using green organic light emitting diodes (OLEDs). Expression of genes and proteins related to proliferation, migration, and adhesion of hASCs irradiated with OLED light was analyzed through various tests to verify tissue regeneration ability. In vivo models were used to analyze wound healing effects of irradiated hASCs. As a result, not only proliferation, migration, adhesion, and expression of growth factors, but also wound healing effects of irradiated cells were enhanced compared to untreated cells. In addition, this study determined molecular mechanisms that stimulate hASCs by using light of a specific wavelength. In conclusion, green OLED light irradiation improved therapeutic efficacy of hASCs, leaving room for development of treatment methods using them.
