제 1장이 연구의 목적은 장용코팅(지연방출)이 필수적으로 요구되어진 기존의 라베프라졸 나트륨 정제와 달리 속방출 제형을 디자인하여 빠른 약효와 위내 안정성을 지닌 속방 라베프라졸 나트륨 경구 제형을 개발하는 것이었다. 라베프라졸은 흔히 사용되는 proton pump inhibitor 약제지만 산성 매질에서 불안정하여 장용캡슐 혹은 장용정형태로만 제조되어왔고 속방형 제형설계는 매우 제한되었었다. 본 연구에서는, “라베프라졸을 내핵으로 함유하고 주성분의 위 내 pH환경에서 안정성을 향상시키기 위해 탄산수소나트륨을 외층부로 함유하는” 속방형 유핵정을 제조하여, pharmaceutical dosage form의 안정성 및 약동학적 프로파일을 연구하였다. 그러나 PPI주성분의 위내 환경 pH에서의 용출 안정성 및 수반되는 문제들의 해결하기 쉽지 않았기 때문에, 우리는 QbD접근법을 적용하여 제형설계를 디자인하고 최적화하였다. In vivo pharmacokinetic 연구에서, 새롭게 제조된 속방 유핵정은 기존 지연방출형 라베프라졸 정제와 비교하였을 때, AUClast는 동일하였으나 최고 혈중농도 도달시간(Tmax)은 최대 7배 이상 빨라지는 것을 확인하였다. 제 2장연구배경 : 텔미사르탄 및 로수바스타틴 칼슘의 고정용량 복합 이층정제는 고혈압 및 고지혈증 치료에 사용된다. 미국약전 (USP; united states pharmacopoeia)의 분석방법과 두 성분에 대한 동시분석 방법이 존재하지만 특이성, 정확성 및 정밀성을 확립하는데 어려움이 있었다. 따라서 제약산업에 적용될 수 있는 새로운 분석방법의 필요성이 강조되고 있다. 목적 : 고혈압 및 고지혈증 치료제로서 텔미사르탄 및 로수바스타틴 칼슘의 고정용량 복합 이층정에 대한 용출샘플의 정량분석을 위한 고성능 액체 크로마토그래피의 분석방법을 개발하고 검증하고자 한다. 방법 : 개발된 분석방법은 USP Category Ⅰ에 따라 검증되었다. 용출시험에 대한 정량분석의 유효성은 구성요소에 대한 분석의 시스템 적합성, 특이성, 선형성, 정확성, 정밀도 및 용액 안정성을 기반으로 검증되었다. 결과 : 개발된 분석방법은 선택적이고 정확했다. 텔미사르탄 및 로수바스타틴 칼슘의 retention time은 각각 8.1분과 2.8분 이었고, 시스템적합성은 각각 0.217과 0.17이었다. 두 성분 모두 10-120% 농도에서 선형성을 나타냈다. 정확도와 정밀도는 허용가능한 범위 내에 있었으며, 용출액 내네서 약물은 장기간 안정하였다. 또한 부형제와 주성분간의 상호작용은 발생하지 않았으며 분석에서 피크간섭은 관찰되지 않았다. 결론 : 텔미사르탄 및 로수바스타틴 칼슘의 이층정에 대한 정량분석은 정확하고 선택적이며 정확했다. 따라서 이 방법은 텔미사르탄 및 로수바스타틴 칼슘제제의 산업적 품질관리에 사용하는데 권장될 수 있다.
Chapter 1 The aim of this study was to develop immediate-release oral rabeprazole sodium tablets with rapid efficacy and gastric stability. In contrast, conventional rabeprazole sodium tablets require an enteric coating. Rabeprazole sodium is a commonly prescribed proton pump inhibitor but it is unstable in acidic solutions. Hence, it has been manufactured and supplied only in enteric-coated tablet form. Immediate-release formulations of this drug are very limited. In this study, we applied the quality by design approach to formulate and optimize an immediate-release dry-coated tablet containing rabeprazole sodium as an inner core and an outer sodium bicarbonate layer stabilizing the active pharmaceutical ingredient at gastric pH. We also investigated the stability of the pharmaceutical dosage form and its pharmacokinetic profile. In vivo pharmacokinetics confirmed that the immediate-release rabeprazole/sodium bicarbonate dry-coated tablet had an AUCt that was bioequivalent to the existing delayed-release rabeprazole tablet but its Tmax was up to seven times faster.Chapter 2Background: Telmisartan and rosuvastatin calcium fixed-dose combination (FDC) double-layer tablet is used for hypertension and hyperlipidemia treatment. Owing to the critical problem in establishing the specificity, precision, and accuracy of the USP analysis method and simultaneous analysis method for both components, an analysis method that could be applied to researchers in the pharmaceutical industry is urgently needed. Objective: To develop and validate a reversed phase-high-performance liquid chromatography (HPLC) method for the quantitative analysis of dissolution samples of telmisartan and rosuvastatin calcium fixed-dose combination (FDC) double-layer tablets, as a hypertension and hyperlipidemia treatment. Method: The developed analysis method was validated according to united states pharmacopoeia (USP) Category I requirements. The validity of the quantitative assay of the dissolution test solution was determined based on the system suitability, specificity, linearity, accuracy, precision, and solution stability of the assay for the active pharmaceutical ingredients (API). Results: The developed method was precise and selective. The retention times of telmisartan and rosuvastatin calcium were 8.1 min and 2.8 min, respectively, and systemsuitability was 0.217 and 0.17, respectively. Both APIs showed linearity at 10-120% concentration. The accuracy and precision were also within acceptable limits. The APIs in the dissolution samples were stable for a long time, no interaction occurred between the excipients and APIs, and no interference was observed in the analysis. Conclusion: The quantitative analysis of the FDC double-layered formulation of telmisartan and rosuvastatin calcium was accurate, selective, and precise. Therefore, this method can be recommended for use in the industrial quality control of telmisartan and rosuvastatin calcium formulations.
