Tumor suppressor p53 is the most frequently mutated or lost protein in various tumor patients, therefore, it is also most interested and studied protein through decades for scientists. Steady and well-controlled function of p53 is important for inhibition of tumorigenesis, growth of tumor, and cell death. Up to recently, MDM2 and ARF have been known for p53 regulator. MDM2 negatively functions as a E3 ligase ubiquitin initiator for p53 degradation. On the contrary, ARF is positive regulator for p53 through the physical binding with MDM2, followed by preventing the function of MDM2. Recent data indicate that ARF controls nucleo-cytoplasmic shuttling and nucleolar targeting of Mdm2. However, still now, How or Where ARF interact with MDM2 is controversial topic because of their different main location, nucleolus and nucleoplasm, respectively. Nucleophosmin is as multifunctional nucleolar protein that functions for cellular growth and proliferation. Recently, several reports have shown that Nucleophosmin targets ARF into nucleolus, and inhibit its function as a p53 positive regulator. However, Even though several studies determined that Nucleophosmin regulates localization of ARF, mechanism of ARF movement is totally unfounded. In this study, we investigated that GLTSCR2 physically interacts with Nucleophosmin. Under cellular stress condition, GLTSCR2 induced translocation of Nucleophosmin into nucleoplasm, followed by exclusion of ARF, which is isolated from nucleoplasm by Nucleophosmin. We also determined that GLTSCR2 induced nucleoplasmic ARF functions as a tumor suppressor in both p53 dependent and independent manner. This suggest that GLTSCR2 is a tumor suppressor protein.
p53은 50% 이상의 암 환자에게서 소실되거나 변이가 존재하는 중요한 암 억제 유전자로서, 현재까지 가장 많은 연구가 이루어지고 있는 단백질이다. 암의 생성과 성장, 억제에 있어서 p53의 정상적인 기능이 매우 중요하며, 다양한 연구를 통하여 p53의 변이가 암을 유발하는 것이 증명되었다. 이러한 p53의 조절인자로서, 현재까지 MDM2와 ARF가 밝혀져 있다. MDM2는 p53의 분해를 촉진하여, p53의 암 억제 유전자로서의 기능을 저해하며, 그에 따른 암 생성을 유발하는 것으로 밝혀졌다. 또한, ARF는 이러한 MDM2와 결합함으로써, MDM2의 p53에 대한 저해작용을 방해하고, 그에 따라, p53의 기능을 유지 및 증가시키는 조절인자로 인식되어지고 있다. 그러나, 이러한 ARF와 MDM2의 상호작용에 있어서, 두 단백질의 세포 내에서의 주 위치가 각각 핵인과 핵질로 서로 상이함으로써, 지금까지 어떻게 두 단백질이 만나고 작용하는지에 대한 논리적 설명의 필요성이 대두되었다. Nucleophosmin은 핵인에 주로 존재하는 다양한 기능을 가진 단백질로서, 세포의 성장, 분열, 그리고 세포사멸의 방지를 위한 역할을 하는 것으로 알려져 있다. ARF와의 관계에 있어서 Nucleophosmin은 ARF와 핵인에서 결합하며, 결합하지 못한 ARF는 핵질로 방출되어 분해된다는 사실이 밝혀졌다. 이는 Nucleophosmin이 ARF를 nucleoli로 유도하고, p53에 대한 ARF의 기능을 억제한다는 것을의미하지만, ARF를 핵인으로부터 핵질로 유도하여 분해되는 것이 아닌 암 억제유전자로서의 기능을 가능하게 하는 mechanism에 대한 연구는 지금까지 이루어지지 않았다. 이번 연구에서는, GLTSCR2가 Nucleophosmin의 binding partner로써 다양한 세포에 대한 자극에 반응하여 protein binding하는 Nucleophosmin을 핵인으로부터 핵질로 이동시키고, 동시에 ARF 또한 핵질로 이동시키는 것을 확인하였다. 그와 더불어, GLTSCR2에 의하여 핵질로 이동한 ARF는 분해되지 않고, Mdm2와의 반응이 증가하는 것을 확인함으로써, 기존의 Nucleophosmin의 감소에 의한 ARF의 핵질로의 이동과는 차이가 있는, 암의 억제를 위한 중요한 현상이라는 점을 확인하였다. 이러한 현상은, 암세포에서의 실험을 통하여 세포주기의 정지와, 세포의 사멸을 유도하는 것 또한 확인하였다. 이번 연구를 통하여 밝혀진, GLTSCR2에 의한 ARF의 조절 기능이 앞으로 p53에 대한 연구와 암의 억제에 대한 연구에 있어서 매우 중요할 것이라 사료되며, GLTSCR2에 대한 연구가 아직 많이 이루어지지 않음으로써, GLTSCR2의 조절과 기전에 관련하여, 많은 연구가 진행되어져야 할 것이다.
