Poorly regulated inflammation is believed to be the most predominant element which could result in a wide variety of diseases including atopic dermatitis, whose prevalence is consistently increasing in both children and adults throughout the world. Pear pomace containing peel, seeds, and core, is the main waste in the manufacturing of pear products. In this present study, we aimed to explore the anti-inflammatory effects of pear pomace ethanol extract (PPE) both in vivo and in vitro. In the in-vivo experiment, the NC/Nga mice were applied with 2, 4-dinitrochlorobenzene (DNCB) to induce atopic dermatitis-like skin lesions. After the successful induction of atopic dermatitis, different concentrations of PPE (200 and 400 mg/kg body weight) were administrated by gavage once daily for four weeks. In relative to the DNCB control group, PPE significantly reduced the scratching behaviors, improved the skin symptoms, and decreased the ear thickness. More importantly, immunoglobulin E (IgE), being one of the authentic prognostic indicators of atopic dermatitis, was significantly decreased by 400mg/kg PPE. Additionally, the expression of the inflammation marker gene inducible nitrite oxide synthase (iNOS) was reversed after the supplement of PPE. Furthermore, PPE treatment inhibited the DNCB-induced receptor for advanced glycation end products (RAGE), extracellular signal-regulated kinase (ERK) 1/2, and nuclear factor kappa B (NF-κB). On the other hand, in the in-vitro experiment, lipopolysaccharide (LPS) was used to induce inflammation in the RAW 264.7 macrophages. Without affecting cell viability, the pretreatment of PPE (100 and 250 ug/ml) significantly inhibited the LPS-induced overproduction of nitrite oxide (NO) and the enhanced expression of iNOS and cyclooxygenase (COX)-2. Moreover, the phosphorylation of ERK1/2 and the activation of NF-κB were also suppressed by PPE. From these results, we proposed that PPE has the inhibitory effects on inflammation and its anti-inflammatory effects were partly through the NF-κB pathway. Collectively, PPE might be served as an anti-inflammatory candidate to treat inflammation disease like atopic dermatitis.
잘못 조절 된 염증은 아토피 성 피부염을 비롯한 다양한 질병을 일으킬 수있는 가장 중요한 요소로 여겨지며, 전염병은 전 세계적으로 어린이와 성인 모두에서 꾸준히 증가하고 있다. 껍질, 씨 및 중핵을 포함하는 배의 찌꺼기는 배 제품 제조의 주요 폐기물이다. 본 연구에서는 체 외 (in vivo) 및 체외 (in vitro)에서 pear pomace ethanol extract (PPE)의 항 염증 효과 연구에 초점을 두었다. 체내 실험에서 NC / Nga 쥐에게 2,4- 디 니트로 클로로 벤젠 (DNCB)을 도포하여 아토피 성 피부염과 유사한 피부 병변을 유도하였고, 아토피 성 피부염이 성공적으로 유도된 이후에 PPE (200 및 400 mg / kg 체중)의 다른 농도를 4 주간 1 일 1 회 위관 영양법으로 투여했다. DNCB 대조군에 비해 PPE는 쥐가 긁는 행동을 현저히 줄이고 피부 증상을 개선하며 귀 두께를 감소 시켰다. 더욱 중요한 것은 아토피 성 피부염의 확실한 예후 지표 중 하나 인 면역 글로불린 E (IgE)가 400mg / kg PPE만큼 현저히 감소한 것이다. 또한 염증 마커 유전자 inducible nitrite oxide synthase (iNOS)의 발현은 PPE 보충 후에 역전되었다. 또한, PPE 처리는 진행된 당화 산물 (RAGE), 세포 외 신호 조절 키나아제 (ERK) 1/2 및 핵 인자 카파 B (NF-κB)에 대한 DNCB 유도 수용체를 억제 하였고, 한편으로는 체내 실험에서 리포 폴리 사카 라이드 (LPS)를 사용하여 RAW 264.7 대 식세포에서 염증을 유도 하였다. 세포 생존 능력에 영향을 미치지 않고, PPE (100 및 250 ug / ml)의 전처리는 LPS에 의해 유발 된 아질산염 (NO)의 과잉 생산과 iNOS 및 cyclooxygenase (COX) -2의 발현을 유의하게 억제 하였고, ERK1 / 2의 인산화와 NF-κB의 활성화는 PPE에 의해 억제되었다. 이러한 결과로부터 우리는 PPE가 염증에 대한 억제 효과를 가지며 그 항 염증 효과가 부분적으로 NF-κB 경로를 통한 것이라고 알게 되었다. PPE는 총체적으로 아토피 성 피부염과 같은 염증 질환을 치료할 수있는 항염증제 후보 물질로 사용될 수 있다.
