To maintain freshness of fruits and vegetables, proper storage and package techniques, such as temperature and humidity control, hypobaric packaging, modified atmosphere packaging, and active packaging, are being applied. In addition to these technologies, other pre-harvest and post-harvest treatments are being investigated for further effect of freshness keeping. Pre-storage UV light treatment on fresh produce is known to inactivate the contaminated microorganisms, activate the defense system and delay the ripening extending the shelf life. As UV light emitting diode (LED) becomes available at relatively low price recently, continuous or intermittent UV treatment during chilled storage is possible in the container or package. This study tried in-situ LED UV treatment onto the fresh produce stored under refrigerated container in order to see its potential in the fresh produce storage and further optimize its application conditions. In the first part of the study, a container system equipped with UV-LED was fabricated for storing cherry tomatoes under computer-controlled conditions of intermittent on-off cycles (4 repeated cycles of 1 hour on/1 hour off). Wavelength (365 and 405 nm) and physical location of the LED (2 and 5 cm above fruit) were studied as variables affecting the respiration, ethylene production and quality preservation of the fruits at 10 and 20℃. 365 nm wavelength gave much higher radiation intensity than 405 nm, and intensity on surface decreased in inverse proportion to square of distance from LED. When compared to non-irradiated control, UV-LED irradiation decreased the respiration by 5-10% at 10℃ while there was no obvious effect at 20℃. Ethylene production was reduced when the fruits were placed at 5 cm distance, while there was no significant difference from control at 2 cm location. The reduction of ethylene production at 5 cm was more pronounced at 20℃. UV-LED irradiation was shown to have delayed increase or lower concentration in carotenoids compared to control treatment. Any negative effect of UV-LED irradiation on ascorbic acid content and firmness was not observed. In the next step, effect of UV-LED irradiation on microbial growth on solid media as a model for food was investigated. To simulate food container conditions, acrylic boxes with three UV-LEDs placed equidistantly on the underside of the top cover were constructed and controlled automatically (on/off time and sequence). Three nutrient agar plates inoculated with microbial strains were placed under the conditions of the LED irradiation inside the box. Plates were irradiated at 2 cm and 5 cm height for 30 minutes. Five microbial strains were tested for their sensitivity to irradiation by UV-LED: Escherichia coli, Bacillus cereus, Bacillus subtilis, Pseudomonas fluorescens and Saccharomyces cerevisiae. 280 nm UV LED showed the greatest microbial suppression during the given storage time and was able to suppress the outgrowth of 4 bacterial strains and 1 yeast at their optimal growth temperatures. No difference in microbial growth was observed between the 2 and 5 cm locations. UV-LEDs of 280 nm have demonstrated their potential for use in food storage systems to suppress microbial spoilage. As the last part, effect of in-container UV LED irradiation on quality preservation of shredded carrots was investigated in the air and modified atmosphere (MA) conditions. Two sets of experiment with Escherichia coli inoculation and with natural microbial flora in the air (two 30 minute on-off cycles of 1 diode/dm2 per day at a location above 2 cm) showed clear and significant effect of the UV LED irradiation on suppression of microbial growth: 280 nm was the most effective by maintaining the lower microbial count by at least 0.5 log(CFU/g) throughout the 6 day storage. The carotenoid contents of shredded carrots subjected to UV LED treatment at 365 and 405 nm in the air were higher than that of control shredded carrots. In the MA condition of O2 of 1.2-4.3% and CO2 of 8.4-10.6% being indifferent with LED wavelengths, 280 nm UV LED irradiation was also effective in inhibiting microbial growth. While there was observed no difference in carotenoids content between untreated and UV LED-treated shredded carrots in MA, UV LED irradiation at 365 and 405 nm was slightly better in DPPH radical scavenging activity. Conclusively use of UV LED in storage container and package has potential to give the benefits of preserving the microbial and nutritional qualities of fresh produce. Condition of proper application to specific commodity and storage needs to be set up by further studies.
현재 과일과 야채의 신선도를 유지하기 위하여, 적정 온도 및 습도를 유지함과 아울러 포장방법으로서 감압포장, 환경기체조절포장, 선도유지제포장 등 여러 포장기법이 적용되고 있다. 본 연구에서는 이러한 여러 수확 후 처리방법과 포장기법에 더욱 선도유지의 효과를 얻기 위하여 빛 에너지를 이용하고자 하였다. 포장 용기 내에 자외선 LED를 장착하여 신선농산물에 조사함으로써 저장성을 향상시키고자 하였다. 본 연구의 첫 부분으로서 자외선 LED를 장착한 포장용기시스템을 제작하였으며, 노트북 컴퓨터와 연결하여 작동 제어가 가능하고, 조사 위치에 따라 강도 조절이 가능하도록 하였다. 자외선 LED의 강도는 5 cm에 비해 2 cm에서 3~4배 정도 높았으며, 동일한 조사 위치에서 365 nm에서가 405 nm보다 약 10배 정도의 높은 강도를 보였다. 1 L 용량의 밀폐 유리용기 시스템 내에서 자외선 LED로 조사된 방울토마토의 호흡은 20℃에 비해 10℃에서 호흡 감소 효과가 나타났다. 파장간의 차이는 미미하나 10℃에서 365 nm가 405 nm에 비해서 작은 차이지만 호흡감소효과가 우수하게 나타났다. 10℃에서 자외선 LED에 의한 호흡감소효과는 5∼10%의 범위에 있었다. 에틸렌 생산에서는 조사 거리 5 cm와 20℃에서 에틸렌 생산 속도가 분명히 낮아지는 경향이다. 방울토마토에서 자외선LED 조사는 호흡과 용기 내 에틸렌 축적을 억제할 수 있는 가능성을 보였으나, 온도조건 및 조사 위치에 따라 그 효과는 달랐다. 자외선 LED 조사에 따라 측정된 경도, ascorbic 함량, 카로티노이드 함량과 같은 화학적 물리적 품질의 변화에는 부정적 영향은 없는 것으로 보였다. 자외선 LED 조사에 의한 신선농산물의 생리적 변화에 이어서 미생물 성장억제에 대한 자외선 LED의 효과를 조사하였다. 식품 용기와 비슷한 조건의 아크릴박스를 제작하여 박스 상단에 균주를 도말한 plate를 두고 아래쪽에 자외선 LED를 장착하여 2 cm와 5 cm 거리에서 30분간 조사하였으며 사용된 균주는 Escherichia coli, Bacillus cereus, Bacillus subtilis, Pseudomonas fluorescens, Saccharomyces cerevisiae이었다. 280 nm 자외선 LED의 조사는 시험균주들의 최적 성장온도에서 각 균주의 성장을 억제시키는데 크게 효과적이었다. 280 nm 자외선 조사는 다른 파장(365, 405 nm)의 조사보다 아주 분명한 미생물 성장 억제효과를 보여주었다. 마지막 부분에서는 이전 실험의 연장으로 절단 당근에 자외선 LED를 조사하였을 때 미생물 품질변화와 함께 카로티노이드 함량과 같은 화학적 품질 변화를 알아보고자 하였다. 먼저 E. coli가 접종된 절단 당근과 접종되지 않은 절단당근을 대기 조건으로 유지된 밀폐되지 않은 플라스틱 용기에서 자외선 LED 조사된 조건으로 10℃에서 저장하여 실험하였다. E. coli가 접종된 절단 당근과 접종되지 않은 절단당근에서 280 nm 자외선 조사 처리를 한 경우 다른 파장(365 nm, 405 nm)을 조사했을 때 보다 더 큰 미생물 성장억제능력을 보여 가장 효과적이었다. 카로티노이드 함량 변화의 경우 405 nm 파장의 자외선 LED 조사가 대조구에 비해 높은 함량을 보였고 365 nm 파장의 조사 또한 대조구에 비해 약간 증가한 경향을 보였다. 또한 일반적 처리과정을 거친 절단당근을 플라스틱 밀폐용기에 넣어 변형기체(modified atmosphere, MA, 3일 이후에 O2 1.2-4.3%와 CO2 8.4-10.6%)조건으로 포장된 상태에서 자외선 LED를 조사했을 경우의 품질변화(총균수, 카로티노이드, DPPH 소거능)를 측정 평가하였다. 카로티노이드 함량에서는 처리구간의 유의적인 차이는 없었지만 자외선 LED 조사가 절단당근의 품질에 부정적인 영향을 주지는 않았다. 앞의 경우와 마찬가지로 MA 포장조건에서 또한 280 nm 자외선 LED 조사가 미생물 성장억제에 효과적인 것으로 나타났다. 항산화 활성에서는 각 처리구간의 큰 차이가 있지는 않았지만 자외선 LED 조사가 항산화 활성을 약간 증가 시키는 것으로 나타났다.
