Silver nanoparticles (AgNPs) are used in a number of consumer products that directly contact the skin. In this study, we investigated toxicities induced by dermal exposure to AgNPs of varying sizes in male SKH1-hairless mice. The dorsal skins of 6-week-old hairless mice were treated with 5-10, 100, and 160 nm silver nanoparticles at 1, 10, or 100 mg/kg body weight per day for 1 month, and blood, urine and organ distributions of AgNPs were then measured by ICP-MS(inductively coupled plasma mass spectrometry). In addition, the long-term effects of exposure to AgNPs on hematology and blood biochemistry were investigated.Silver concentrations increased in a dose dependent manner in all organs and body fluids examined after treatment with 5-10nm AgNPs. In decreasing order, concentrations followed the pattern liver > kidney > heart > skin > brain > RBC > urine > serum. After treatment with 100 nm or 160 nm AgNPs, silver concentrations also increased dose-dependently in organs and body fluids. Furthermore, silver concentrations were found to be significantly dependent on particle size (5 nm > 100 nm > 160 nm). Blood biochemistry analysis showed that GOT(glutamic oxaloacetic transaminase), ALP(alkaline phosphatase), TG(triglyceride), and BUN(blood urea nitrogen) values were significantly elevated in serum after treatment with 5-10nm AgNPs at 100mg/kg. These results suggest that AgNPs, depending on particle size and exposure level, are absorbed by skin and may produce hepatotoxicity and/ or nephrotoxicity in vivo.
항균, 소독, 항혈소판제 역할을 하는 은(Ag)은 나노 크기로 제조되어 의료기기, 화장품, 창상 드레싱제 (wound dressings), 의복, 치약, 섬유유연제 등 다방면으로 사용되고 있다. 은 나노물질은 크게 작업상의 노출, 소비자로의 노출, 그리고 환경상의 노출 경로가 있으며 인간에게 노출될 수 있는 경로는 경구, 피부, 흡입, 섭취 등이 있다. 화장품, 샴푸, 바디로션, 섬유 등 은나노 물질이 소비자의 피부에 노출될 수 있는 상품들이 많아지고 있음에도 불구하고 아직 나노물질에 대한 허용농도 범위와 라벨링 기준 등이 구체적으로 정해져 있지 않아 나노물질을 사용한 상품들에 대한 규제를 할 수 없는 실정이다. 현재 은나노물질의 피부 독성연구가 미미한 상태이고 나노물질의 NOAEL(No Observed Adverse Effect Level: 독성이 관찰되지 않는 용량)과 LOAEL (Lowest-observed-adverse-effect-level: 최저독성량) 기준치 등 규제치 설정에 노력을 기울여야 할 때이다. 따라서, 본 연구에서는 여러 나노물질 중 은 나노물질의 피부 독성평가(안전성)에 대해서 연구하였다. Hairless mouse의 등 피부 (3cm × 4cm)에 5-10nm, 100nm, 그리고 160nm 크기의 은나노 콜로이드를 1, 10, 100mg/kg 농도로 도포한 후 멸균된 거즈로 고정시켰다. 4주간 은나노를 투여한 후, 마우스를 부검하여 간, 신장, 뇌, 등 피부, 심장을 적출하고, 혈액과 뇨도 채집하여 은 정량 분석을 실시하였다. 4주간 마우스의 몸무게변화량을 측정하고 혈청 생화학적 검사, 조직병리검사, GSH, catalase의 활성 측정 그리고 TEM 과 EDS 분석을 실시하였다. 5-10nm 크기의 고농도군(100mg/kg) 모든 장기에서 은 농도가 유의적으로 증가하였고, GOT, ALP, TG, BUN의 수치가 5-10nm 처리군에서 농도의존적으로 증가하였다. 또한 조직병리검사에서는 5-10nm 고농도군과 100nm 고농도군의 신장 근위곡 세뇨관(proximal convoluted renal tubules)의 세포질에서 액포(vacuoles)가 관찰되었고, 다른 장기에서는 특별한 병변이 관찰되지 않았다. in vitro 상에서 은 나노물질이 세포 내로 침투하는지 관찰하기 위해, hairless mouse 의 등 피부에서 각질세포(keratinocytes)를 primary로 분리하여 은나노를 처리한 후, TEM 분석을 실시하였다. 그 결과, 5-10nm, 100nm, 160nm가 각질세포의 세포질로 침투하였고, 특히 5-10nm는 세포의 핵 내로 침투한 것이 관찰되었다. 그리고 in vivo상에서 은 나노물질이 표피층, 진피층까지 침투하는지 관찰하기 위해서 4주간 은나노를 투여한 피부를 분리하여 TEM 분석을 실시하였다. 모든 크기에서 표피층, 진피층까지 은 나노물질이 침투하였음을 확인하였다. 또한, 간에서의 GSH 활성 측정결과, 5-10nm, 100nm, 160nm 처리군에서 농도의존적으로 감소하였고, 또한 catalase 활성 측정결과, 5-10nm, 100nm, 160nm 처리군에서 농도의존적으로 증가하였고, 5-10nm 중간농도, 고농도군(10, 100mg/kg)에서 유의적으로 증가하였다. 위의 결과들을 보면 은 나노물질 (5-10nm 크기 100mg/kg농도)이 은 피부로 노출되었을 경우에 hairless mouse의 간과 신장에 독성을 유발할 가능성이 있는 것으로 사료된다.
