Aminocoumarin is a class of antibiotics that act by an inhibition of the DNA Gyrase enzyme involved in the cell division in bacteria. They comprise three “classical” compounds, which are similar in structure: novobiocin, clorobiocin and coumermycinA1. They are produced by different Streptomyces strains, exhibit potent activity against Gram-positive bacteria. Aminocoumarin has been introduced into the drug market as an antibiotic for use in humans and recently maybe there are some new therapy functions into people’s attention, such as anti-cancer. Since little is known about the function of aminocoumarins on bone metabolism, I investigate the effect of aminocoumarins on the RANKL-induced osteoclast formation. The multinuclear osteoclasts was differentiated from mouse bone marrow macrophage (BMM) cells in the presence of M-CSF and RANKL. I use coumermycinA1 as one representative reagent of aminocoumarins to further examine the effect of aminocoumarins on the RANKL-induced osteoclast formation in vitro. CoumermycinA1 inhibited RANKL-induced osteoclast formation without cytotoxicity. The mRNA levels of osteoclastogenic markers were decreased with the treatment of coumermycinA1. In consistent with this result, coumermycinA1 induced the disruption of actin rings in mature osteoclasts and suppressed osteoclastic pit formation on dentin slices. In experiments to elucidate its mechanism of action, coumermycinA1 was found to significantly inhibit RANKL-induced gene expression of c-Fos and nuclear factor of activated T-cells (NFATc1), essential transcription factors for osteoclast development. Further study verified that the inhibitory effect of coumermycinA1 is attributable to modulation of signaling pathways of ERK1/2, p38 mitogen-activated protein kinase (MAPK). Therefore, the anti-osteoclastogenic effect of coumermycinA1 is likely to be elicited by interference with RANKL signaling to ERK, p38, which mediate the induction of c-Fos and subsequently that of NFATc1. Furthermore the inhibitory effect of coumermycinA1 on osteoclast formation was partly rescued by retroviral NFATc1 and c-fos over-expression. I used novobiocin to investigate the bone loss in vivo. In accordance with in vitro study, novobiocin treatment reduced LPS-induced osteoclast formation and bone loss in vivo. Taken together these results indicate that aminocoumarins exist a new inhibitory effect on RANKL-induced osteoclast formation in vitro and in vivo and it could be useful for the treatment of bone diseases associated with excessive bone resorption.
Aminocoumarin은 Streptomyces에서 분리된 항생물질로, 박테리아의 세포분열에 관여하는 DNA gyrase를 저해하는 작용을 한다. Gyrase는 DNA복제 시 분리시킨 양쪽 DNA가닥을 회전시키는 데 관여하는 DNA 선회효소로 복제에 필수적인 효소이며, 불활성화되면 염색체 이상을 유발하여 세균의 증식을 저해하게 된다. Aminocoumarin계열 화합물로는 novobiocin, clorobiocin, 그리고 coumermycin A1이 있다. Aminocoumarin은 항생제로 사용되고 있으며, 최근 연구에서는 항암작용과 같은 새로운 기능이 규명되고 있다. 하지만, amonicoumarin의 골대사에 대한 작용기전은 거의 알려진 바가 없다. 따라서, 본 연구에서는 aminocoumarin 계열 항생물질인 courmermycin A1과 novobiocin 을 이용하여 RANKL에 의해 유도되는 파골 세포 형성에 aminocoumarin이 미치는 작용을 조사하였다. 실험 결과, coumermycin A1은 세포독성을 보이지 않는 농도에서 파골 세포의 분화를 억제하는 것으로 나타났으며, 또한 actin ring의 형성과 골 흡수에 의해 형성되는 pit의 생성도 억제하는 것으로 나타났다. 그리고, 파골 세포의 형성과 활성에 관여하는 유전자들의 발현 또한 coumermycin A1에 의해 억제되는 것으로 나타났다. 이러한 효과는 파골 세포 형성에 중요한 역할을 하는 전사인자인 c-fos와 NFATc1의 발현에서도 확인할 수 있었으며, c-fos와 NFATc1을 과잉발현 시켰을 때, coumermycin A1의 파골 세포 형성 억제 효과가 회복되는 것을 확인하였다. 이는 coumermycin A1이 RANKL에 의한c-fos와 NFATc1의 발현을 저해함으로써 파골 세포 형성을 억제함을 나타낸다. 또한novobiocin을 이용한 개체수준의 실험에서도, LPS에 의한 파골 세포 형성과 골 손실에 대한 저해 효과를 확인할 수 있었다. 이상의 결과는 aminocoumarin이 파골 세포의 형성을 저해하여 골 손실을 억제함을 시사하며, 골 흡수의 증가로 나타나는 골 질환 치료제로서의 가능성도 제시한다.
