Endogenous Retroviruses (ERVs) are unique exterior elements which have been originated from ancient infectious exogenous retroviruses. Among several ERV families, ERV9 is a low repeated family of HERV elements, which has close to 50 members and at least 4000 solitary long terminal repeats (LTRs). Those LTR elements have potential of transcription regulatory factors by providing promoter activity, various transcription factor binding sites, and cryptic splicing sites. Using bioinformatic and evolutionary analyses, molecular features of LTR12 of ERV9 family are investigated in relation to functional genes. As LTR12 fused genes, some of the LTR12 members are served as either promoter or exon. Alternative transcripts derived from LTR12 appeared in several human tissues. The LTR12 fused CHM gene showed different expression patterns between normal and adjacent tumor tissues. In RAE1 gene, LTR12 showed strong promoter activity in HEK293 cell line. Integration event of the LTR12 of ERV9 family during primate evolution suggests that most of LTR12 of ERV9 family have been entered into the African great apes lineage. Taken together, the LTR12 of ERV9 family may contribute to the gene regulation and transcript diversification for host genome.
내생 레트로 바이러스인 ERVs(endogenous retroviruses) 의 기원은 고대 외생 레트로 바이러스가 생식 세포 내로 감염되어 시작하였다 다양한 내생 레트로 바이러스 패밀리 중, ERV9 패밀리는 인간 내생 레트로바이러스 중 적은 수로서 인간 유전체에 약 4000 개의 단독 LTR(Long Terminal Repeat) 을 가지고 있다. 이러한 LTR 은 프로모터의 역할, 전사 조절 인자의 접합 부위 제공, 그리고 스플라이싱 서열을 제공하는 등 다양한 전사 조절자로서의 역할을 한다. 따라서 본 연구에서는 생물 정보학적 및 진화적 접근을 통하여 인간 유전체에서 LTR12의 역할을 규명하였다. LTR12가 포함된 10개의 유전자를 결정하여 그 유전자 내에서 LTR의 역할을 프로모터와 엑손화로 구분하였다. LTR12에 의해 만들어진 전사체들은 인간 조직에서 다양한 발현 양상을 나타내었으며 TBX1 유전자를 제외한 나머지 9개의 유전자에서 LTR12에 의한 인간 특이적인 전사체의 탄생을 확인하였다. 또한 LTR12에 의하여 만들어진 CHM 유전자의 isoform b 전사체는 정상 조직과 암 조직에서 각각 다른 발현량을 보였다. RAE1 유전자에는 LTR12이 프로모터 영역에서 작동하고 있었는데 이들의 프로모터 활동성이 아주 강력한 것으로 확인하였다. 영장류의 진화 과정 동안 이들의 특징은 10개의 유전자가 각기 다른 시기에 LTR12 삽입되어 영향을 미쳤다는 것을 확인 할 수 있었으며, 특히 오랑우탄과 고릴라의 분기에 삽입이 많았다. 결과적으로 이러한 공통 조상으로부터 진화 과정 동안 LTR12의 삽입은 유전자의 조절 및 발현 그리고 전사체의 발생 등에 영향을 미친 것을 확인 할 수 있었다. 또한 LTR12가 삽입된 전사체의 조직 특이적인 발현 패턴은 이후 암을 확인하는 바이오 마커로서의 역할도 기대해 볼 수 있을 것이며 조직 특이적인 질병 및 유전자 발현 연구에 큰 도움이 될 것으로 생각한다.
