Antipsychotic drugs are widely used to alleviate a number of psychic disorders. Treatment with second-generation antipsychotics has been associated with weight gain and diabetes mellitus, but the molecular mechanism of their action has been poorly recognized. Leptin and insulin are key hormones involved in the regulation of energy balance and glucose homeostasis. Development of resistance to the action of these hormones, which can occur with obesity and inflammation, appears to have a prime role in the pathogenesis of obesity and type 2 diabetes. In this study, I investigated effects of risperidone on insulin-induced PKB phosphorylation and leptinstimulated STAT3 phosphorylation in human neuroblastoma SH-SY5Y cell line. Risperidone specifically increased suppressor of cytokine signaling 3 and 6 (SOCS3 and SOCS6). And also, induction of SOCS3 and SOCS6 require cAMP-dependent PKA activity, which activated extracellular signal-related kinase (ERK), and leading to ERK-dependant up-regulation of SOCS3 and SOCS6 proteins. SOCS3 down regulated receptor activation of JAK2/STAT3 leptin signaling pathway. SOCS3 and SOCS6 inhibited insulin-induced PKB phosphorylation. Taken together, these data suggested that risperidone activated PKA and induced expression of SOCS3 and SOCS6. Then inhibited phosphorylation of STAT3 and PKB, which may be an important mechanism of risperidone caused weight gain in schizophrenic patients, and also provided a new role of risperidone for some syndromes therapy includes insulin and leptin resistance.
항정신병약은 정신장애를 완화하는데 광범위하게 사용되고 있 다. 제2세대의 항정신병약의 치료는 체중 증가와 제2형 당뇨병을 유발하고 있는 부작용이 있지만 그들의 분자적 작용기전은 아직 알려지지 않았다. 랩틴과 인슐린은 에너지 평형과 포도당의 항상 성을 유지시키는 중요한 호르몬이다. 이러한 호르몬의 작용에 있 어 저항성의 증가는 비만과 염증을 유발하는 것으로 나타나고, 이 것이 비만과 제2형 당뇨병을 일으키는 중요 요인으로 작용한다.본 연구에서는 인체 신경암세포인 SH-SY5Y 세포에서 유도성 PKB 인산화와 랩틴 유도성 STAT3 인산화에서의 Risperidone의 효 과를 연구했다. Risperidone은 사이토카인 신호전달 3과 6의 억제인 자 (SOCS3/SOCS6)를 현저하게 증가시킴을 관찰하였다. SOCS3/SOCS6의 증가는 cAMP 의존 단백질 키나아제 (PKA)의 활 성이 요구되고, 외부신호 전달 관련 단백질 키나아제 (ERK)의 활 성화를 유도했으며 이는 ERK 의존적 SOCS3/SOCS6의 상승 조절 을 의미하게 된다. SOCS3는 JAK2/STAT3의 수용체 활성을 억제하 였다. 또한 SOCS3와 SOCS6는 인슐린 유도성 PKB 인산화를 억제 시켰다.결론적으로 일련의 실험결과는 Risperidone이 PKA를 활성화 시키고, SOCS3와 SOCS6의 발현을 유도하여 STAT3와 PKB의 인산 화를 억제함을 의미한다. 이것은 아마도 정신분열증환자에서 Risperidone의 약물부작용인 체중 증가의 중요한 원인이 되고, 인슐 린과 랩틴 저항성을 포함한 여러가지 질병 치료요법에서 Risperidone의 새로운 역할을 보여준 것이다
