ERRγ는 Ligand가 없는 상태에서도 전사 활성을 증가 시키는 핵수용체 대가족 집단의 일환이다. 그러나 ERRγ 에 의한 정확한 유전자 조절의 기작은 잘 아직 까지 잘 알려지지 않고 있다. 본 연구자는 고아핵수용체 ERRγ 가 DAX-1 promoter를 조절하고 다시 DAX-1에 의해서 ERRγ의 활성이 억제 됨을 연구하였다. Mouse DAX-1 gene promoter를 이용한 Serial deletions mutant 실험 결과 ERRγ 에 반응하는 부위가 DAX-1 promoter 의 -129 and -121 bp 와 -334 and -326 bp 사이에 위치함을 확인하였다. EMSA와 ChIP assays 실험 결과는 ERRγ 가 DAX-1 promoter에 직접적으로 결합함을 보여주었다. 그리고 Site-directed mutagenesis 실험 결과 ERR 반응 부위중 ERRE1이 ERRE2 보다 중요함을 확인하였으며 ERRE2는 Human promoter에 보존되어 있지 않음을 확인하였다. 유방암 세포주인 MCF-7 세포에서 ERRγ Adenovirus를 통하여 과발현 시켰을 경우 DAX-1의 유전자 발현이 현저히 증가함을 확인하였다. 또한 Yeast two-hybrid, GST-pull down assays를 통하여 DAX-1이 ERRγ에 직접적으로 결합함을 확인하였고 전사 활성도를 억제 하는 것을 발견하였다. 그리고 이 결합은 ERRγ의 AF-2 domain에 의존적임을 확인하였다. In vitro competition assays 결과 DAX-1 이 PGC-1a와 ERRγ 의 결합을 AF-2 binding domain 의존적으로 전사 활성도를 억제함을 확인하였다.ERRγ는 전사조절 인자로 작용하기 때문에 ERRγ의 목적 유전자를 규명 하는 것이 ERRγ의 기능을 이해하는데 중요할 것으로 사료되어 본 연구자는 293T 과 INS-1 세포주에 ERRγ의 Adenoviral infection으로 과발현을 유도한 후 DNA Microarray를 수행하였다. DNA Microarray 실험 결과 Polo-like kinase 2 (Plk2)의 유전자 발현이 293T 과 INS-1 세포주에서 현저히 증가함을 확인하였다. 또한 Northern blot 실험결과 여러 암세포주에서 ERRγ가 Plk2유전자의 발현을 증가시킴을 보여주었다. 게다가 ERRγ가 Plk2 gene promoter를 활성화 시킴을 확인하였다. Serial deletions과 Mutational analysis를 통하여 ERRγ에 의한 Plk2 gene promoter 반응부위가 -2327 와 -2229 bp 사이 그리고 -441 and -432 bp 부위에 위치함을 결정하였으며 EMSA 와 ChIP assays 결과 ERRγ 가 Plk2 promoter에 직접적으로 결합함을 확인하였다. 최근에 보고되어진 ERRγ agonist, GW4716 역시 Plk2 유전자의 발현을 증가시켰으며 Plk2 promoter 를 활성화 시킬 수 있었다. ERRγ가 과발현 되었을 경우 Nocodazole에 의한 민감도가 암세포에서 감소하였으며 Cell cycle 동안 S기를 증가시킴을 확인하였다. 이는 ERRγ가 암세포주에서 세포의 성장에 영향이 있음을 보여주는 결과이다.본 연구자는 ERRγ 에 의한 DAX-1 의 유전자 발현이 Autoregulatory loop을 통하여 조절되며 Plk2가 ERRγ의 목적 유전자임을 규명하였다. 이는 향후 ERRγ의 기능을 연구하는데 많은 도움을 줄 것으로 사료된다.
Estrogen receptor-related receptor gamma (ERRγ/ERR3/NR3B3) is a member of the nuclear receptor superfamily that activates transcription in the absence of ligand. However, the detailed mechanism of gene regulation by ERRγ is not fully understood. In this study I have found that the orphan nuclear receptor ERRγ activates the DAX-1 promoter, which, in turn, represses transactivation by ERRγ. Serial deletions of mouse DAX-1(mDAX-1) gene promoter have revealed that the region responding to ERRγ is located between -129 and -121 bp and -334 and -326 bp. Gel shift assays and chromatin immunoprecipitation (ChIP) assays demonstrated that ERRγ binds directly to the mDAX-1 promoter. Site-directed mutagenesis results demonstrated that ERRE1 (-129 to -121 bp) is more important than ERRE2 (-334 to -326 bp) which is not conserved in the human DAX-1 promoter. In addition, adenovirus-mediated overexpression of ERRγ induced DAX-1 gene expression in MCF-7 breast cancer cells that co-expressed ERRγ and DAX-1. Moreover, yeast two-hybrid and GST-pull down assays demonstrated that DAX-1 physically interacted with ERRγ and inhibited ERRγ transactivation, and that this interaction was dependent on the AF-2 domain of ERRγ. In addition, in vitro competition assays showed that DAX-1 inhibited PGC-1a mediated ERRγ transactivation, via competition between these two factors for the AF-2 binding domain. To identify putative target genes of ERRγ, I performed DNA microarray analysis of gene expression following adenovirus-mediated overexpression of ERRγ. Polo-like kinase 2 (Plk2) was significantly up-regulated by overexpression of ERRγ. Northern blot analysis showed that overexpression of ERRγ induced Plk2 expression in cancer cell lines. ERRγ activated the Plk2gene promoter, and deletion and mutational analysis of the Plk2 promoter revealed that the ERRγ-response region is located between nucleotides (nt) -2327 and -2229 and -441 and -432 (relative to the transcriptional start site at +1). Electrophoretic mobility shift assay (EMSA) and chromatin immunoprecipitation (ChIP) analysis demonstrated that ERRγ binds directly to the Plk2 promoter. The ERRγ agonist GW4716 also activated the Plk2 promoter and induced Plk2 expression. Overexpression of ERRγ in the presence of the mitotic inhibitor nocodazole significantly decreased apoptosis,and induced S-phase cell cycle progression through the induction of Plk2 expression.In summary, I suggest a novel autoregulatory loop that controls DAX-1 gene expression by ERRγ and Plk2 is a novel target of ERRγ. This investigation will provide new insights into understanding the function of ERRγ.
