An Efficient and Stable Method for the Transformation of Heterogeneous Genesinto Cephalosporium acremonium Mediated by Agrobacterium tumefaciens

A transformation system mediated by Agrobacteriumtumefaciens is routinely used for the genetic engineering ofplants. Here, we report an efficient and stable methodfor transformation of heterogeneous genes into an industrialCephalosporium acremonium by using a similar transformationsystem established in plants. Both the phleomycin-resistantgene and vgb gene were used as screening markers to confirmthe success of transformation by either Southern hybridizationor PCR amplification. It was found that acetosyringone (AS)was necessary only for protoplast transformation and theheterogeneous genes transferred were integrated into the genomeof C. acremonium. The transformation efficiency obtainedwith this system was much higher than the conventionaltechniques used for transformation of C. acremonium.
