目的 探讨维药榅桲多糖对3T3-L1小鼠胚胎成纤维细胞增殖及胰岛素抵抗的作用.方法 培养3T3-L1脂肪细胞,通过噻唑蓝比色(MTT)法观察不同浓度榅桲多糖对细胞增殖的影响;通过油红O染色法并测定甘油三酯(TG)的含量,评价前脂肪细胞是否诱导为成熟脂肪细胞;采用地塞米松为诱导剂建立3T3-L1胰岛素抵抗细胞模型,分别给予不同浓度榅桲多糖进行干预后,以葡萄糖氧化酶法检测培养液中葡萄糖残留量.结果 与正常对照组比较,榅桲多糖能明显抑制3T3-L1脂肪细胞增殖;油红O染色法判定前脂肪细胞成功诱导为成熟脂肪细胞,TG含量测定结果表明,与正常对照组相比成熟3T3-L1细胞中脂质含量增多( P<0.01);在3T3-L1胰岛素抵抗细胞模型中,与正常对照相比榅桲多糖可明显增加葡萄糖利用率( P<0.05).结论 榅桲多糖具有抑制脂肪细胞增殖的作用并可明显改善地塞米松诱导的胰岛素抵抗.
Objective To study the effect of Cydonia oblonga Mill.polysaccharide on proliferation and insulin resistance in 3T3-L1 cells.Methods The effect of polysaccharide in Cydonia oblonga Mill was observe on proliferation of 3T3-L1 cells by MTT colorimetric assay;The differentiation degree of adipocytes was analyzed by oil red O staining and detection of triglyceride;The insulin resistance model of adipocyte was induced by dexamethasone, the glucose concentration in cell culture medium was determined after drug treatment.Results Compared with normal control group, Cydonia oblonga Mill. polysaccharide could inhibit proliferation;The 3T3-L1 cells were differentiated into mature adipocytes which could be observed by oil red O staining and the TG concentration was increased compared with normal group ( P<0.01);The 3T3-L1 cells model of insulin resistant was successfully established and Cydonia oblonga Mill.polysaccharide could improve glucose utilization compared with normal group ( P<0.05).Conclu-sion Cydonia oblonga Mill.polysaccharide could inhibit adipocyte proliferation and ameliorate the level of IR of 3T3-L1 induced by dexamethasone significantly.
