Antibiyotiklere direnç gösteren fırsatçı patojen bakterilerin giderek yaygınlaşması sonucu kronik enfeksiyonların sayısı artmakta ve insan sağlığı ciddi sorunlarla karşılaşmaktadır. Hastalık Kontrol ve Korunma Merkezleri (CDC) tarafından yayınlanan 2019 yılı raporuna göre, ABD'de her yıl 2.8 milyondan fazla antibiyotiğe dirençli enfeksiyon vakalarının meydana geldiği ve bunun sonucunda 35.000'den fazla insanın hayatını kaybettiği bildirilmektedir. Staphylococcus aureus (S. aureus), dünyada gıda ve yoğun bakım ünitelerinden kaynaklı olarak toplum ve hastane yoluyla enfeksiyonlara yol açan önemli mikroorganizmalardan biri olduğu bilinmektedir. Quorum Sensing (QS) mekanizmasının, virülans üretimi ve biyofilm oluşumu yoluyla savunma mekanizmalarının aktivasyonuna yanıt olarak gen ekspresyon modelini değiştiren bakteriyel bir iletişim süreci olduğu literatürde bildirilmektedir. QS sistemlerinin doğal bileşiklerle inhibisyonu, geleneksel antibiyotiklere alternatif bir yaklaşım olabilmektedir. Liken özütlerinden biyoaktif metabolit üretme potansiyeli, yeni doğal ilaçlar için dikkat çekmektedir. Bu çalışmada, S. aureus'a karşı liken özütlerinin QS ve biyofilm inhibisyon potansiyeli araştırılmıştır. Bursa–Alaçam yöresinden liken örnekleri toplanmıştır ve aseton çözücüsü ile ekstre edilmiştir. Belirli konsantrasyonlarda test edilmiştir. S. aureus P2 Lac–gfp ve P3 Lac–gfp biyosensör suşları ile S. aureus ATCC25923 yabanıl tip suşu, QS ve biyofilm deneyleri için kullanılmıştır. Biyosensör suşlarda anti–QS ve yabanıl suşta anti–biyofilm için inhibisyon oranları incelendiğinde ortak olarak Usnea chaetophora, Hypogymnia physodes ve Pseudevernia furfuracea liken özütlerinin etkili olduğu görülmüştür. Liken özütleri gibi doğal bileşiklerin taranması, gelecekteki QS inhibitör ilaçları için fırsatlar sağlayabileceği düşünülmektedir. As opportunistic pathogenic bacteria resistant to antibiotics become more widespread, the number of chronic infections is increasing, and human health is facing serious problems. According to the 2019 report published by the Center for Disease Control and Prevention, it is reported that more than 2.8 million antibiotic–resistant infections occur in the USA every year, and more than 35.000 people die as a result. Staphylococcus aureus (S. aureus) is known to be one of the important microorganisms that cause infections in the community and hospital, originating from food and intensive care units in the world. S. aureus is one of the important microorganisms that cause community and hospital infections. It is specified in literary sources that QS mechanism is a bacterial communication process that leads to changes in gene expression such as producing virulence factors and forming biofilm as a response to the activation of the organism's defense mechanism. Blocking the QS mechanism with natural sources can be an alternative to antibiotics. Metabolite productions from lichens are noteworthy for new natural remedies. In this study, the QS and biofilm inhibition potential of lichen extracts against S. aureus was investigated. Lichen samples were collected from Bursa region and extracted with acetone solvent. Tested at specific concentrations. S. aureus biomonitor strains P2 Lac–gfp and P3 Lac–gfp and wild type strain S. aureus ATCC25923 were used for QS and biofilm experiments. When the inhibition rates for anti–QS and anti–biofilm were examined, Usnea chaetophora, Hypogymnia physodes and Pseudevernia furfuracea lichen extracts were found to be effective. It is envisaged that natural compounds such as lichen extracts may be opportunities for new QS inhibitory agents.
