본 연구에서는 이팝나무(Chionanthus retusa) 잎 추출물을 사용하여 항산화와 주름개선 효과를 측정하고, 에멀션 제형의 화장품을 제조하여 안정성(pH, 색도, 점도, patch test 및 관능 평가)을 평가하였다. 이팝나무 잎의 total phenolic compounds 함량은 열수 및 에탄올 추출물에서 각각 17.85, 36.66 mg GAE/mL의 함량을 나타내었다. 이팝나무 잎 추출물의 DPPH 라디칼 소거능은 각각 89.20, 91.52%의 효과를 나타내었다. Elastase 억제 효과는 에탄올 추출물 600 μg/mL 농도에서 99.63%의 저해율을 나타내었으며, collagenase 저해 효과는 에탄올 추출물 600 μg/mL 농도에서 59.03%의 저해율을 나타내어 매우 우수한 주름생성 억제 효능을 나타내는 것을 확인할 수 있었다. 이팝나무 추출물을 활용한 화장품을 4주간 저장하며 pH, 색도, 점도를 측정한 결과, pH, 점도에서는 차이를 보이지 않았다. 색도의 경우 추출 시료를 첨가한 농도에 따라 전체적으로 변화가 있었으며, 4주 동안 저장기간에 따른 변화는 열수 추출물에서만 L값이 2%의 농도에서 감소하였고 a값은 전체적으로 증가하였다. 화장품의 patch test에서는 화장품으로 인해 일어날 수 있는 피부 알레르기 반응은 나타나지 않았다. 화장품의 관능 평가에서는 화장품의 색, 향, 발림성, 흡수도, 전체적인 기호도에서 이팝나무 잎 추출 동결건조물 첨가 농도가 높아질수록 색과 전체적인 기호도에서 남녀 모두 평가 점수가 낮아졌으며, 흡수도의 경우 에탄올 추출물로 제조한 화장품에서 점수가 높았다. 발림성의 경우 에탄올 추출물 화장품에서만 여성 평가자의 점수가 증가하였으며, 그 외의 향과 발림성에서는 차이를 보이지 않았다. 이상의 결과에 따라서 이팝나무 잎 추출물의 페놀성 화합물 함유량과 항산화 활성, 주름개선 효과 및 화장품 안정성을 확인하여 기능성을 가지는 화장품 제조 원료로 개발 가능성을 확인하였다.
In this study, the anti-oxidant and anti-wrinkle activities and emulsion cosmetic-assosiated stability characteristics (pH, colorimetric, viscosity, patch test, and sensual properties) of Chionanthus retusa leaf (CRL) extracts were evaluated. Total phenolics contents of CRL hot water and 60% ethanol extracts were 17.85 and 36.66 mg GAE/g, respectively. At 200 μg/mL hot water and ethanol extracts had 2,2-diphenyl-1-picrylhydrazyl radical scavenging activities of 89.20 and 91.52%, respectively, and at 600 μg/mL had elastase inhibitions of 79.42 and 99.63%, and collagenase inhibitions of 32.29 and 59.03%, respectively. These results confirmed the anti-oxidant and anti-wrinkle properties of the active ingredients in CRL extracts. After four weeks of storage, CRL extracts had no effects on the pH, color, and viscosity of 10 types of cosmetics patch testing for skin reactions revealed no abnormality. Sensory evaluations on the color, scent, spreading, absorption, and overall preferences of cosmetics showed men and women scored the hot water extract better for color and overall preference but found the ethanol extract better for skin absorption. The study indicates that CRL extracts have anti-oxidant activities and stabilities suitable for use in cosmetics.
