BACKGROUND:: Establishing an effective and complete animal model is a precondition for further research of spinal cord injury (SCI), which can provide a basis for SCI treatment. OBJECTIVE:: To establish a stable animal model of acute complete SCI in rats. DESIGN, TIME AND SETTING:: A contrast control animal experiment was performed at the Center of Animal Experiment in College of Pharmaceutical, Shihezi University between November 2007 and October 2008. MATERIALS:: Thirty healthy, Wistar rats were randomly divided into the control group (n=6), and the experimental group (n=24). METHODS:: Rats in the experimental group were exposed processus spinosus and vertebral plate of T8-T12, and cut processus spinosus, and vertebral plate of T9-10 of corresponding spinal cord were served as injured region. The weight drop device was designed with the cerebrum three-dimensional positioning indicator based on Allenʼs model. All of experimental animals were attacked T10 segment (15 g×20 cm=2.94×10 N). Be sure that the integrity and continuity of the dura in the experimental group were not damaged. There was no attack in the sham operation group. MAIN OUTCOME MEASURES:: The motor function of both hind limbs were evaluated by oblique-plate test and BBB scores at 2, 4, and 8 weeks after operation. The pathological change of myeloid tissue was observed by hematoxylin-eosin staining. RESULTS:: Rats in the sham operation group could walk after reviving, and the angle of oblique-plate test was more than 70°, with 21 scores. The micro-structure of spinal cord was normal. Rats in the experimental group were paralyzed completely after operation. Two rats showed spastic paralysis, and 5 rats showed different degree of self-mutilation. The angle of oblique-plate test was less than 30° with lower than 10 scores at weeks 2, 4, and 8 after operation. With the time increasing, some of rats had reflex activity to stimulation, but without active movement. The local inner-structure of spinal cord was damaged severely. CONCLUSION:: An animal model of spinal cord injury with stable and complete can be established by attract with 2.94×10 N, which can ensure the integrity of spinal dura after operation.Zhang H, Liu WG, Huang J, Li KX, Xi SS, Zhang FJ, Li DZ.
