전 세계적으로 뇌졸중으로 인해 고통 받는 사람이 점차적으로 증가하는 추세이다. 그 중에서도 뇌혈관이 막혀서 문제가 생기는 허혈성 뇌졸중의 비율이 점차 높아지고 있다. 그래서 허혈성 뇌졸중에 관한 연구가 많이 진행되고 있다. 하지만 허혈성 뇌졸중 동물 모델이 다소 부정확하고 문제점이 있다고 언급되고 있다. 본 논문에서는 실시간 모니터링 시스템을 이용하여 보다 정확하고 재현성 강한 허혈성 뇌졸중 동물 모델의 개발에 대하여 연구하였다. 실험 결과 기존의 사용하던 11VO 모델에서 1) 흡입 마취제로의 변화, 2) 2단계 시술시간의 변화로 기존의 방법과 같은 실험 결과를 얻을 수 있었다. 가장 문제가 되던 실험 시간을 연장 시킬 수 있게 되었다. 본 논문을 통하여 기존의 취약점을 보안한 발전된 모델을 확인 할 수 있었고, 이것을 통하여 앞으로 모니터링 후, 염색을 통한 형태학적 변화의 관찰과 행동 실험을 통한 변화를 관찰할 수 있게 되었다. 그래서 앞으로 허혈성 뇌졸중을 연구하는데 중요한 보탬이 될 것이라 사료된다.
This study examined the compare with original 11 vessel occlusion (11VO) and modified 11VO model. Modified 11VO method was change of surgical operation time (original: one day operation, modified: one day- occlusion of basilar artery and next day- another operation) and anesthetic (original: used chloral-hydrate, modified: one day use chloral-hydrate and next day use anesthetic breathing) from original method. The real-time extracellular glutamate concentration was measured using a microanalysis biosensor and the CBF was monitored by laser Doppler flowmetry. Neuronal cell death in the hippocampal region was observed 72 hr after ischemia by several stains (Nissl, DAPI, NeuN, and cleaved caspase3). The modified 11VO group showed not different pattern compare to original 11VO. Level of glutamate release and change of CBF pattern was similarly observed each group during the ischemic and reperfusion episode. Our staining results revealed a significant decrease in neuronal cell death in the hippocampus in the ischemia group compared to the sham group. We suggested that modified 11VO model could be help to research of ischemic stroke.
