Neurodegeneration, a term that refers to the progressive loss of structure and function of neurons, is a feature of many neurodegenerative diseases (NDD) such as Alzheimer’s disease (AD), Parkinson’s disease and Huntington’s disease. Pathophysiologically, among several etiological factors, aging and neuroinflammation are well known major risk factors in various NDD. Mechanistically, both factors play vital role in dysregulation of c-Jun N-terminal kinase (JNK)-a stress kinase belong to a subfamily of mitogen-activated protein kinase. Although JNK regulate various important brain functions. However, abnormal activation of JNK/c-Jun pathway ties with several neuropathological features associated with NDD including initiation of mitochondrial apoptotic pathway, neuroinflammation, synaptopathy, cognitive deficits/memory impairment and amyloidogenesis associated with AD-a NDD. On opposite in terms of treatment, inhibition of JNK/c-Jun pathway has been suggested as a worthwhile approach to combat ageing and inflammation mediated NDD. In this regard, in addition to different strategies (JNK knockout techniques, chemical compounds and cell-permeable peptide), considerable attention has also been paid to natural sources in research studies. Therefore, in the present study, we first established animal models of neurodegeneration; a D-galactose (D-gal)-induced aging and lipopolysaccharide (LPS)-mediated neuroinflammation by using different neurotoxic agents (D-gal and LPS) and then examined the neuroprotective effects of natural compounds (Glycine (Gly) against D-gal; and Vanillic Acid (V.A) against LPS) through behavioral tests and biochemical and immunofluorescence assays. Our in-vivo results indicated that both neurotoxins (D-gal or LPS) induced oxidative stress-mediated neurodegeneration primarily by upregulating phospho-JNK (p-JNK) levels. However, their respective cotreatment either with Gly or V.A reversed neurotoxic effects by down regulating p-JNK levels in mice brain. Noticeably, this inhibitory effect on p-JNK was verified by both molecular docking approach and in-vitro study (HT22 murine hippocampal neuronal cell lines were treated with or without a p-JNK-specific inhibitor (SP600125)); indicated that Gly bound to p-JNK protein and inhibited its function and its downstream singling. For instance, we found that Gly reversed D-gal-induced neuroapoptosis by significantly reducing the protein expression levels of proapoptotic markers (Bax, cytochrome c, cleaved caspase-3, and cleaved PARP-1) and increasing the protein expression level of the antiapoptotic protein Bcl-2. In addition, we also found that Gly or V.A alleviated neurotoxins-mediated neuroinflammation by inhibiting gliosis (astrocytosis and microgliosis) and reducing the protein expression levels of various pro-inflammatory cytokines (IL-1βeta, TNFα and COX-2). Aside from this, we noted that V.A co-treatment significantly attenuated LPS-induced increases in the expression of AD markers, such as β-site amyloid precursor protein–cleaving enzyme 1 and amyloid-β. Furthermore, we also found that both Gly and V.A reversed neurotoxins-induced synaptic dysfunction by upregulating the expression of memory-related presynaptic protein markers (synaptophysin, syntaxin, and a postsynaptic density protein) and markedly improved behavioral measures of cognitive deficits; as accessed by Morris Water Maze and Y-Maze. Taken together, we propose that these underlying potent neuroprotective effects of Gly and V.A against neurotoxins-induced neurotoxicity might be due to deactivation of JNK signaling pathway. However, more future studies are highly encouraged to further evaluate their precise underlying molecular mechanism in neuroinflammatory and various age-related neurodegenerative disorders.
뉴런의 구조와 기능의 점진적 상실을 일컫는 용어인 신경퇴화는 알츠하이머병, 파킨슨병, 헌팅턴병 등 많은 신경퇴행성 질환의 특징이다. 여러 가지 병리학적 요인 중 노화와 신경염증은 다양한 퇴행성 뇌질환의 주요 위험인자로 잘 알려져 있다. 기전적인 관점에서 두 인자는 모두 mitogen-activated 단백질 kinas 계열의 c-Jun N-terminal kinase(JNK)의 조절 장애에 중요한 역할을 한다. 속한다. JNK/c-Jun 신호전달 경로의 비정상적 활성화는 미토콘드리아 세포사멸 유발, 신경 염증, 시냅스 손상, 인지결손/기억장애 및 알츠하이머성 치매와 관련된 아밀로이드 생성을 포함한 여러 신경병리학적 특징과 밀접한 관련이 있다. 치료제 개발의 측면에서, JNK/c-Jun 신호전달 경로의 억제는 노화 및 염증 매개 퇴행성 뇌질환을 방지하기 위한 유망한 있는 접근법으로 제안되었다. 이와 관련하여 연구자들은 다양한 전략(JNK knock-out 기법, 화학 화합물 및 세포 투과성 펩타이드)외에도, 쳔연 유래 치료 후보물질 등 다양한 연구를 수행했다. 본 연구에서는 먼저 D-galactose(D-gal) 유도 노화 및 lipopolysaccharide(LPS) 매개 신경염증 동물 모델을 확립한 후 행동 검사와 생화학 및 면역 형광 검사를 통해 천연 화합물(Glycine, Gly, Gilly)의 신경보호 효과를 확인하였다. 생체내 결과에 따르면 두 신경독소 compounds 모두 주로 인산화 된-JNK(p-JNK) 발현량을 증가시킴으로써 산화성 스트레스 매개 신경퇴행을 유발하였다. 그러나 Gly, V.A(D-gal에는 Glycine (Gly) 처리,LPS에 Vanillic Acid (V.A) 처리) 처리시 생쥐 뇌에서 p-JNK 발현량을 감소시켜 신경독성을 완화시켰다. 유의미하게, p-JNK에 대한 이러한 억제 효과는 분자 도킹 접근법과 시험관내 연구(HT22, 쥐 해마 신경 세포주에서 p-JNK 특이 억제제(SP600125) 처리됨)에 의해 검증되었으며, Gly결합이 p-JNK 단백질과 그 기능과 하위 신호 독성유발 단백질 발현량을 억제했음을 나타냈다. 예컨대, Gly는 세포사멸 단백질(Bax, cytochromec, cleaved caspase-3, cleaved PARP-1)의 단백질 발현 수치를 유의미하게 감소시키고 항 세포사멸 단백질 Bcl-2의 단백질 발현량을 높여 D-gal 유발 신경사멸을 완화시켰다. 게다가, 본 연구에서는 또한, Gly과 V.A가신경독성에 의해 유발된 신경염증을 완화(성상세포와 미세아교세포 활성화 억제) 및 다양한 염증성 사이토카인(IL-1βeta, TNFα, COX-2)의 단백질 발현 수치를 감소시키는 것을 확인하였다. 또한 V.A 처리시 LPS처리에 의해 증가된 β-site amyloid precursor protein–cleaving enzyme 1 과 Amyloid-β와 같은 알츠하이머성 치매 관련 단백질의 발현량을 유의미하게 감소시켰다. 또한 Gly, V.A 처리시 신경독성에 의해 감소된 시냅스 단백질(synaptophysin, syntaxin, PD95 등) 발현량을 증가시켰고 Morris Water Maze와 Y-Maze행동 실험에 기억력 장애가 현저하게 개선되었음을 확인하였다. 결론적으로 본 연구를 통해 Gly와 V.A의 잠재적인 신경보호 효과를 검증하였으며 그 과정에서 신경독성에 의해 유발된 JNK 신호 경로를 비활성화 하는 것을 확인하였다. 후속적으로 신경염증 및 다양한 연령 관련 신경퇴행성 질환에서 정확한 기본 분자 메커니즘을 추가로 평가하기 위해 향후 연구가 필요할 것으로 사료된다.
