Backgrounds: Recent studies have shown that not only neurons but astrocytes contain a considerable amount of GABA, which can be released and activate the receptors responsive to GABA. GABA from reactive astrocytes has been implicated in epilepsy, sleep, memory and cognition. The mechanism underlying the astrocytic release of GABA in depression is poorly understood. The purpose of this study is to test whether gliotransmitters from astrocytes may play a role in etiology of depressive-like symptoms.Methods: After rat recombinant IL-1β was infused into 3rd ventricle (100 ng, ICV) or paraventricular nucleus (10 ng, i.PVN) of the hypothalamus (PVN), the rats were tested for depressive-like behaviors in elevated plus maze, open field test and tail suspension test. Expression of glial fibrillary acidic protein (GFAP) and γ-aminobutyric acid (GABA) were investigated using the immunofluorescence. Next, the systemic or local pharmacologic glial inhibition (L-α-aminoadipate; L-AAA) or bestrophin-1 channel blocker (5-Nitro-2-(3-phenylpropylamino) benzoic acid; NPPB) was pretreated before IL-1β ICV or i.PVN injection and depressive-like behaviors and GABA and GFAP expression in the PVN were measured. Finally, extracellular GABA release was measured using in vivo microdialysis in order to examine the role of astrocytic GABA in IL-1β induced depressive behaviors.Results: I.c.v infusion of IL-1β (100ng) induced depressive-like behaviors and activated the glial fibrillary acidic protein (GFAP) in PVN and hippocampal dentate gyrus (DG). However, astrocyte inhibition by systemic L-α-aminoadipate (L-AAA; 0.5, 5mg/rat, s.c.) reduces depressive behaviors and the GFAP expression in the PVN. Injection of IL-1β (10ng) into the PVN produced markedly depressive behaviors and release GABA from GFAP. Local injection of L-AAA (250nmol/μL/bilateral) or blockade of Best1 channel by systemic 5-Nitro-2-(3-phenylpropylamino) benzoic acid (NPPB; 40mg/kg, i.p. or 300nmol/μL/bilateral), specific Best1 channel blocker, decreased depressive behaviors, but not changed the GABA expression in the PVN. Finally, IL-1β (10 ng/rat) injection into the PVN release GABA measured by in vivo microdialysis, and treatment of L-AAA or NPPB decreased IL-1β-induced gliotransmitter GABA release.Conclusion The present results demonstrated that release of gliotransmitter GABA in the brain may modulate the interleukin 1β-induced depressive responses. These results suggest that selective inhibition of astrocyte or astocytic GABA release may serve as an effective therapeutic strategy for treating depression.
배 경: 최근의 연구들은 신경세포뿐만 아니라 성상세포도 감마-아미노부틸산 (GABA) 의 상당한 양을 포함하고 있다고 알려져 있다. 이러한 성상세포는 GABA를 합성 및 분비 할 수 있고 GABA에 반응하는 수용체들을 활성화 할 수도 있다. 반응성 성상세포로부터 분비된 GABA는 간질, 수면, 기억과 인지에 관여 하지만 우울증에서 성상세포성 GABA의 분비에 관한 기본 메커니즘은 매우 적은 부분만이 이해되고 있다. 따라서 본 연구의 목적은 성상세포로부터 분비되는 신경아교전달물질이 우울증과 같은 증상에서 병인학적 역할을 수행 하는지 규명하고자 한다.방 법: 재조합된 인터루킨-1베타를 제 3뇌실 (100 ng) 또는 시상하부의 실방핵에 (10 ng) 주입한 후, 우울 행동을 확인하기 위해 실험동물들은 십자형 미로, 오픈 필드 검사, 그리고 생쥐 꼬리 현수법을 수행 하였다. 또한, 뇌내 스트레스 반응을 관장하는 실방핵에서 아교섬유산성단백질 (GFAP) 과 GABA의 발현을 면역형광염색을 이용해서 확인하였다. 다음으로 인터루킨-1베타를 제 3뇌실 또는 실방핵에 투여하기 전에, 성상세포만 특이적으로 억제하는 L-α-aminoadipate (L-AAA) 또는 성상세포에서 GABA분비한다고 알려진 Bestrophin-1 채널을 선택적으로 차단하는 5-Nitro-2-(3-phenylpropylamino) benzoic acid (NPPB)를 전체 혹은 국소적으로 투여하고 우울 행동과 GFAP-GABA의 발현 측정하였다. 마지막으로, 인터루킨-1베타로 야기된 우울 행동에서 성상세포성 GABA의 역할을 확인하기 위해 세포 외 GABA분비는 생체 내 미세투석법을 이용하여 측정하였다.결 과: 제3뇌실에 인터루킨-1베타의 투여는 우울행동을 증가시켰고, 실방핵에서 GFAP활성화 시켰다. 그러나, 전체적인 L-AAA (0.5, 5mg/rat, s.c.) 투여에 의한 성상세포의 활성의 억제는 우울행동을 감소시켰으며, 실방핵에서 GFAP 발현을 억제하였다. 실방핵에 인터루킨-1베타의 국소적인 투여는 현저한 우울행동과 GFAP-GABA의 발현을 나타냈다. 국소적으로 L-AAA (250 nmol/μL/bilateral) 또는 NPPB (300 nmol/μL/bilateral)를 투여한 결과, 두 가지 모두 우울행동을 감소 시켰다. 하지만, 실방핵에서 GFAP와 GABA의 발현은 변화가 없었다. 마지막으로 실방핵 내에 인터루킨-1베타를 주입 후 분비된 GABA의 양을 측정하기 위해 실방핵에서 생체 내 미세투석법을 실시한 결과, 인터루킨-1베타의 실방핵 내 국소 투여는 GABA의 분비를 증가시켰으며, L-AAA 또는 NPPB의 주입 후에는 인터루킨-1베타로 유도된 성상세포성 GABA의 분비가 감소되었다.결 론: 본 연구는 뇌내 신경아교전달물질 GABA의 분비가 인터루킨-1베타로 유도된우울 반응을 조절할 수도 있다는 것을 보여주었다. 이러한 결과는 선택적인 성상세포의 활성억제 또는 성상세포성 GABA 분비의 억제는 우울증 치료에 효과적인 치료 전략으로서 역할을 할 수 있다고 제안한다.
